西藏移居內(nèi)地的藏族和漢族不適癥狀大學(xué)生血清蛋白表達(dá)分析

【摘要】:背景隨著世居高原人群往返于高原與平原之間愈發(fā)頻繁,高原低氧人群對(duì)平原富氧環(huán)境的適應(yīng)逐漸引起人們的重視。目的基于血清蛋白組學(xué)對(duì)移居內(nèi)地的藏、漢族不適癥狀大學(xué)生的血清蛋白表達(dá)進(jìn)行分析,以期為西藏移居內(nèi)地的藏、漢族大學(xué)生因適應(yīng)不良而引起的疾病提供簡(jiǎn)單易行的、精確的早期診斷指標(biāo),為其臨床治療提供分子靶標(biāo)。方法分別選取2013年10—11月、2014年10—11月、2015年10—11月西藏民族大學(xué)符合納入標(biāo)準(zhǔn)的藏、漢族不適癥狀大學(xué)生32例為研究對(duì)象。按照研究對(duì)象民族,將其分為藏族組和漢族組,各16例。收集研究對(duì)象血清樣本,采用弱陽離子磁珠(MB-WCX)捕獲血清蛋白多肽,采用基質(zhì)輔助激光解吸電離飛行時(shí)間質(zhì)譜(MALDI-TOF-MS)技術(shù)進(jìn)行質(zhì)譜分析并比較兩組血清蛋白表達(dá)水平。結(jié)果單樣本質(zhì)譜分析結(jié)果顯示,3次重復(fù)取樣,藏族組同一個(gè)體分子量為1 000~10 000 m/z的血清蛋白多肽曲線基本趨于一致,漢族組得出相似結(jié)果;藏族組與漢族組血清蛋白表達(dá)水平存在顯著差異。整體樣本質(zhì)譜分析結(jié)果顯示,藏族組與漢族組分子量為1 000~10 000 m/z的血清蛋白表達(dá)水平存在顯著差異;對(duì)其蛋白多肽成分分析,則呈現(xiàn)出相互聚集、較少重合的現(xiàn)象;藏族組與漢族組分子量為905.65、1 062.10、1 078.11、2 138.79、2 313.40、4 094.67、4 172.89、4 213.96、5 910.71 m/z的血清蛋白表達(dá)峰值間存在顯著差異(P0.05)。結(jié)論西藏移居內(nèi)地的藏、漢族不適癥狀大學(xué)生血清蛋白表達(dá)存在一定差異,其中分子量為4 094.67、2 138.79、905.65 m/z的血清蛋白表達(dá)差異較大,為進(jìn)一步鑒定和定位差異性蛋白的序列和基因奠定了基礎(chǔ)。
[Abstract]:Background as the commute between the plateau and the plain becomes more and more frequent, the adaptation of the people in the plateau to the oxygen-enriched environment in the plain gradually attracts people's attention. Objective to analyze the expression of serum protein in Tibetan and Han unwell symptom college students based on seroproteomics, so as to provide a simple and easy way for Tibetan students to emigrate to Tibet and Han nationality college students for diseases caused by maladjustment. The accurate early diagnosis index provides the molecular target for the clinical treatment. Methods from October to November 2013, October to November 2014 and October to November 2015, 32 college students with unwell symptoms of Han nationality were selected as subjects. According to the nationalities of the subjects, they were divided into Tibetan group and Han group, 16 cases each. Serum samples were collected and serum protein peptides were captured by weak cationic magnetic beads (MB-WCX). Matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) technique was used to analyze the expression of serum proteins in two groups. Results the results of single sample mass spectrometry showed that the curve of serum protein polypeptide with a molecular weight of 1 000 ~ 10 000 m / z in the Tibetan group was almost the same as that in the Han group, and the same result was obtained in the Han nationality group. There was significant difference in the expression of serum protein between Tibetan group and Han nationality group. The results of mass spectrometry showed that there was a significant difference between Tibetan group and Han group in the expression of serum protein with a molecular weight of 10 000 m / z, and the analysis of protein polypeptide showed a phenomenon of mutual aggregation and less overlap. There was significant difference in the peak expression of serum protein between the Tibetan group and the Han nationality group when the molecular weight was 905.65 (106.102.102.108.1110) 2138.79 (2313.404094.67) and 4172.89 (4213.966) 5 910.71 m / z (P0.05). Conclusion there are some differences in the expression of serum protein between Han Chinese and Tibetan who emigrated to the mainland of Tibet, and there is a significant difference in the expression of serum protein with molecular weight of 4.094.672138.79905.65mz. It provides a basis for further identification and localization of the sequences and genes of differential proteins.
