LC Orexin-A對(duì)肥胖抵抗大鼠自發(fā)活動(dòng)的影響及機(jī)制研究
[Abstract]:Aim: to investigate the effect and mechanism of locus coeruleus (LC) orexin-A on spontaneous activity in obese resistant (OR) rats and to explore whether the orexin regulation of LC is related to obesity resistance. Methods: male SD rats and selectively cultured OR rats were injected with 0.5 渭 L orexin-A or synthetic cerebrospinal fluid (CSF) slowly and evenly. SPA infrared moving-sensing box was used to detect the spontaneous activity of rats. The energy consumption was measured by (SPA), indirect calorimetry and the fat weight and muscle weight were measured by quantitative magnetic resonance body composition analyzer. The degree of obesity in rats was expressed as a percentage of body fat. Results: compared with SD rats, the total weight, fat weight and degreasing weight of OR rats were significantly decreased (P0.05). OR rats were significantly less obese than SD rats (P0.05). OR rats at 3 months of age) showed horizontal activity. The vertical activity increased at 6 months, suggesting that the activity of OR rats increased significantly with age (P0.05). Compared with SD rats, SPA level of OR rats was significantly higher (P0.05), non-resting energy expenditure significantly increased (P0.05), but there was no significant difference between day energy expenditure (P0.05). The results suggested that increased night-time activity in OR rats resulted in increased energy consumption and weight loss. LC injection of orexin-A could significantly increase SPA of OR rats (P0.05) in a dose-dependent manner, but had no significant effect on the feeding intake of OR rats (P0.05). These results suggest that the orexin-A system of LC plays an important role in obesity resistance in OR rats, and this effect may be achieved through its regulation of SPA. Conclusion: orexin-A plays an important role in the spontaneous activity of obese rats, which is related to obesity resistance.
