母代營養(yǎng)過剩對雄性子代能量代謝的影響
[Abstract]:Objective to investigate the effect of maternal excess nutrition on energy metabolism of male offspring. Methods C57BL/6J female rats were randomly divided into high fat diet group and normal diet group, fed with high fat diet or normal diet for 2 months, then mating with normal diet C57BL/6J male rats. Pregnant rats continued to take high fat or normal diet during pregnancy and lactation. After sexual maturation, the male mice of the first generation mate with the normal diet C57BL/6J female mice to obtain the second generation, and the second generation of the male mice after the sexual maturation of the second generation then mate with the normal diet C57BL/6J female mice to get the third generation. The body weight, food intake and random blood sugar of the male mice of the second generation, the second generation and the third generation were measured. The intraperitoneal glucose tolerance test was carried out in the second generation and the second generation of adult male mice, and the wet weight of liver, periepididymal fat and subcutaneous fat were recorded. Serum insulin and hypothalamic energy metabolism related genes were detected in the second generation adult male mice. Results the body weight, food intake, energy intake and random blood glucose of the male mice of the second generation and the second generation were higher than those of the control group (P0.05), but there was no significant difference between the three generations (P0.05). The ratio of periepididymal lipids to lipids, subcutaneous lipids to lipids, livers to lipids and the area under the glucose tolerance curve of adult male mice with over nutrition were higher than those of the control group (P0.05). Serum insulin concentration and hypothalamic neuropeptide Y (NPY), associated protein (AGRP), glycine (GAL), (GAL), cocaine-(CART), in adult male mice of second generation over-fed female mice The expression of SIM1 gene was higher than that of control group (P0.05). Conclusion the effect of high fat diet on the energy metabolism of male offspring can be sustained up to the second generation, resulting in abnormal expression of hypothalamic energy metabolism related genes in the second generation of hypothalamus.
