貝那普利對早期糖尿病腎臟疾病足細胞損傷保護作用的實驗研究

【摘要】:目的探討貝那普利對早期糖尿病腎臟疾病(DKD)足細胞損傷保護作用。方法選取SPF級雄性Wistar大鼠40只,隨機分為正常對照(NC)組、單腎切除(SNE)組、DKD組和貝那普利(Ben)組,每組10只,其中DKD組和Ben組采用60mg/kg STZ腹腔注射建立DKD模型,成模4周后,Ben組予10mg/(kg·d)貝那普利灌胃,DKD組、SNE組和NC組4周后予等量蒸餾水灌胃。檢測各組生化指標(biāo),觀察腎臟組織病理改變及足細胞超微結(jié)構(gòu),免疫熒光檢測腎皮質(zhì)Nephrin和Podocin蛋白表達。結(jié)果Ben組多飲多尿癥狀減輕,第4、8、12周體質(zhì)量均高于DKD組(P0.05);DKD組和Ben組血糖高于NC組、SNE組(P0.05);DKD組Scr和24h尿蛋白定量高于NC組、SNE組,血清白蛋白低于NC組、單腎切除組(P0.05);Ben組Scr和24h尿蛋白定量(81.30±20.46)μmol/L、(190.44±5.10)mg/24h低于DKD組,血清白蛋白(36.47±1.32)g/L高于DKD組(P0.05);DKD組腎組織AT Ⅱ高于NC組(P0.05);Ben組AT Ⅱ(14.60±0.82)pg/ml低于DKD組(P0.05);DKD模型組足突寬度、足突融合率和基底膜厚度高于NC組、SNE組(P0.05);Ben組足突寬度、足突融合率和基底膜厚度分別為(0.41±0.01)μm、(34.20%±6.81%)和(0.40±0.03)μm,低于DKD組(P0.05);Ben組Nephrin和Podocin蛋白表達較DKD組升高。結(jié)論貝那普利對早期DKD足細胞有保護作用,其機制可能與降低腎組織ATⅡ,上調(diào)Nephrin和Podocin蛋白表達有關(guān)。
[Abstract]:Objective to investigate the protective effect of benazepril on (DKD) podocyte injury in early diabetic nephropathy. Methods Forty SPF grade male Wistar rats were randomly divided into normal control (NC) group, single nephrectomy (SNE) group, DKD group and benazepril (Ben) group with 10 rats in each group. DKD model was established by 60mg/kg STZ intraperitoneal injection in DKD and Ben groups. After 4 weeks, Ben group was given 10mg/ (kg d) benazepril, DKD group, SNE group and NC group were given the same amount of distilled water after 4 weeks. The pathological changes of renal tissue and the ultrastructure of podocytes were observed. The expressions of Nephrin and Podocin protein in renal cortex were detected by immunofluorescence. Results the symptoms of polydipsia and polyuria were alleviated in Ben group, and the body mass of DKD group was higher than that of DKD group at the 12th week (P0.05). The blood glucose in); DKD group and Ben group was higher than that in NC group and SNE group (P0.05). The levels of Scr and 24 hours urinary protein in DKD group were higher than those in NC group, while in SNE group, serum albumin levels were lower than those in NC group and single nephrectomy group (P0.05).
