燃煤型氟中毒對(duì)大鼠骺板軟骨組織形態(tài)學(xué)的影響

【摘要】:目的模擬病區(qū)人群復(fù)制燃煤型氟中毒大鼠模型,動(dòng)態(tài)觀察大鼠不同時(shí)期骺板軟骨組織形態(tài)的改變。方法 150只SD大鼠按體重隨機(jī)分為對(duì)照組、中氟組、高氟組、高氟伴低蛋白低鈣組、高氟伴高鋁組,每組30只(雌雄各半),分別于第60 d、第120 d、第180 d處死,取大鼠左膝關(guān)節(jié),觀察大鼠骺板軟骨組織病理學(xué)改變。結(jié)果各染氟組與對(duì)照組相比,骺板邊緣不齊,軟骨細(xì)胞鈣化延遲,增殖層和肥大層細(xì)胞層數(shù)增多;高氟伴低蛋白低鈣組細(xì)胞柱排列紊亂,隨染氟時(shí)間延長,出現(xiàn)大量壞死區(qū);高氟伴高鋁組增殖層軟骨偶爾可見結(jié)節(jié)樣增生;對(duì)于同一實(shí)驗(yàn)組不同時(shí)期比較,隨著大鼠飼養(yǎng)時(shí)間延長,骺板軟骨組織的增殖層細(xì)胞層數(shù)均逐漸降低(P0.05);不同時(shí)期3批大鼠各染氟組骺板增殖層、肥大層的細(xì)胞層數(shù)均多于對(duì)照組,且隨著染氟劑量增高細(xì)胞層數(shù)逐漸增加(P0.05);高氟伴低蛋白低鈣組、高氟伴高鋁組骺板增殖層、肥大層細(xì)胞層數(shù)較高氟組均降低。結(jié)論燃煤型氟中毒大鼠出現(xiàn)了骺板軟骨組織的損傷;肥大層、增殖層細(xì)胞層數(shù)增多;高氟伴低蛋白低鈣組大鼠骺板組織出現(xiàn)大量壞死區(qū);高氟伴高鋁組大鼠骺板組織增厚主要表現(xiàn)為結(jié)節(jié)性增生。
[Abstract]:Objective to simulate the rat model of coal-burning fluorosis and observe the morphological changes of epiphyseal plate cartilage in different periods. Methods one hundred and fifty SD rats were randomly divided into control group, middle fluoride group, high fluoride with low protein and low calcium group, and high fluoride and high aluminum group with 30 rats in each group (half male and female). The rats were killed on the 60th, 120th and 180th day, respectively. The histopathological changes of the rat epiphyseal plate cartilage were observed by taking the left knee joint. Results compared with the control group, the epiphyseal plate margin was irregular, the calcification of chondrocytes was delayed, the number of cell layers in proliferative layer and hypertrophic layer increased in each fluorine-exposed group, and the cell column of high fluorine group with low protein and low calcium was disarranged, and a large number of necrotic areas appeared with the prolongation of fluorine exposure time. Nodular proliferation was occasionally seen in proliferation layer cartilage of high fluoride with high aluminum group. Compared with the same experimental group, the number of proliferation layer cell layer of epiphyseal chondrocytes decreased gradually with the prolongation of feeding time (P0.05). The proliferation layer of epiphyseal plate and the number of cell layer in hypertrophic layer of three groups of rats exposed to fluoride in different periods were all more than those in control group, and the number of cell layers increased gradually with the increase of fluoride concentration (P0.05). The proliferation of epiphyseal plate and the number of mast cell layer in high fluorine group with low protein and low calcium were lower than those in high fluoride group. Conclusion the damage of epiphyseal chondrocytes appeared in the rats with coal-burning fluorosis, the number of cell layers in the hypertrophic layer and proliferative layer increased, and a large number of necrotic areas appeared in the epiphyseal plate of the rats with high fluoride and low protein and low calcium. The thickening of epiphyseal plate in high fluoride with high aluminum group was mainly nodular hyperplasia.
