雙酚A對(duì)高脂飼料小鼠脂肪組織炎癥及Th17細(xì)胞的影響

【摘要】:目的探討雙酚A(BPA)暴露對(duì)高脂飼料小鼠脂肪組織炎癥及Th17細(xì)胞的影響。方法高脂飼料(HFD)喂養(yǎng)的4周齡雄性C57BL/6小鼠,通過飲水?dāng)z入10、100和1000 nmol/L BPA,并設(shè)立正常飼料(ND)和HFD對(duì)照組。每周記錄小鼠體重,BPA暴露8周和16周后處死小鼠,取附睪脂肪墊,HE染色觀察脂肪組織病理學(xué)改變,酶聯(lián)免疫吸附實(shí)驗(yàn)(EILSA)檢測脂肪組織中炎癥因子TNF-α的含量,流式細(xì)胞術(shù)(FCM)檢測脂肪組織中Th17細(xì)胞比例,免疫組化(IHC)檢測脂肪組織中IL-17的表達(dá)。結(jié)果 8周和16周時(shí),與ND對(duì)照組相比,HFD對(duì)照組和HFD飼料BPA暴露組小鼠體重明顯增加,附睪脂肪組織炎性細(xì)胞浸潤增多,TNF-α含量上升,Th17細(xì)胞比例增加,IL-17表達(dá)上調(diào);與HFD對(duì)照組相比,BPA暴露組小鼠附睪脂肪組織炎性細(xì)胞浸潤增多,TNF-α含量上升,Th17細(xì)胞比例增加,IL-17表達(dá)上調(diào),且具有劑量-效應(yīng)關(guān)系;與8周時(shí)相比,16周時(shí)變化更明顯。結(jié)論 BPA暴露可促進(jìn)高脂飼料小鼠脂肪組織炎癥、Th17細(xì)胞比例增加和IL-17表達(dá)上調(diào);而Th17細(xì)胞可在BPA促進(jìn)脂肪組織炎癥中發(fā)揮作用。
[Abstract]:Objective to investigate the effects of bisphenol A (BPA) exposure on adipose tissue inflammation and Th17 cells in mice fed with high fat diet. Methods four week-old male C57BL/6 mice fed with high fat diet (HFD) were fed with drinking water for 10100 nmol/L BPA, and 1000 nmol/L BPA, respectively, and normal diet (ND) and HFD control group were set up. The weight of mice was recorded every week. The mice were killed after BPA exposure for 8 weeks and 16 weeks. The epididymal fat pad was taken, the pathological changes of adipose tissue were observed by HE staining, and the content of inflammatory factor TNF- 偽 in adipose tissue was detected by (EILSA) by enzyme-linked immunosorbent assay (Elisa). The percentage of Th17 cells in adipose tissue was detected by flow cytometry (FCM) and the expression of IL-17 in adipose tissue was detected by immunohistochemical (IHC). Results at 8 and 16 weeks, compared with ND control group, HFD control group and HFD feed BPA exposure group significantly increased their body weight, infiltration of inflammatory cells in epididymal adipose tissue, increase of TNF- 偽 content, increase of Th17 cell proportion and up-regulation of IL-17 expression. Compared with the control group of HFD, the inflammatory cell infiltration of epididymal adipose tissue increased, the content of TNF- 偽 increased, the proportion of Th17 cells increased and the expression of IL-17 increased in BPA exposed group, which had a dose-effect relationship. The changes were more obvious at 16 weeks than at 8 weeks. Conclusion BPA exposure can promote the inflammation of adipose tissue, increase the proportion of Th17 cells and up-regulate the expression of IL-17 in high-fat diet mice, while Th17 cells can play a role in the promotion of adipose tissue inflammation by BPA.
