【摘要】：目的誘導(dǎo)擴(kuò)增BALB/C小鼠滑膜抗原特異性調(diào)節(jié)性T細(xì)胞(antigen specific regulatory T cells,sTregs)的產(chǎn)生,為探索滑膜抗原sTregs在類風(fēng)濕性關(guān)節(jié)炎(rheumatoid arthritis,RA)的治療作用提供實(shí)驗(yàn)基礎(chǔ)。方法體外誘導(dǎo)培養(yǎng)小鼠骨髓源性樹突狀細(xì)胞(DCs)并負(fù)載Ⅱ型膠原蛋白肽,在白細(xì)胞介素-2(IL-2)的協(xié)同作用下,與小鼠脾CD4~+CD25~+自然發(fā)生的調(diào)節(jié)性T細(xì)胞(nTregs)混合培養(yǎng),以刺激產(chǎn)生滑膜抗原sTregs,并用混合淋巴細(xì)胞抑制實(shí)驗(yàn)評(píng)估擴(kuò)增的sTregs抑制CD4~+CD25~-T細(xì)胞增殖的功能是否具有抗原特異性。結(jié)果經(jīng)DCs刺激1周后,小鼠CD4~+CD25~+Tregs擴(kuò)增了6倍,并且增殖的sTregs在混合淋巴細(xì)胞抑制試驗(yàn)中較多克隆Tregs(Poly-Tregs)具有較強(qiáng)的抑制作用(P0.01),顯示抗原特異性抑制的特點(diǎn)。結(jié)論以自身DCs負(fù)載滑膜源性抗原(Ⅱ型膠原蛋白肽)可以從小鼠脾CD4~+CD25~+nTregs中誘導(dǎo)擴(kuò)增滑膜抗原sTregs。
