【摘要】：目的探討蘆丁在肥胖雄性小鼠體質(zhì)量抑制以及對肥胖誘發(fā)的生殖損傷的保護(hù)效果及其機(jī)制。方法將24只雄性ICR小鼠隨機(jī)分成3組,每組8只:正常對照組(ND組,正常飲食),高脂飲食組(HFD組,高脂飲食),蘆丁保護(hù)組(HRU組,高脂飲食+蘆丁)。28 d后,按后續(xù)實驗需要進(jìn)行取材處理。試劑盒檢測生化指標(biāo);睪丸與附睪進(jìn)行石蠟切片,HE染色進(jìn)行形態(tài)學(xué)觀察;統(tǒng)計精子數(shù)、活動度和畸形率。熒光定量PCR檢測相關(guān)基因的表達(dá)水平;Western blot檢測Ucp1蛋白表達(dá)水平。結(jié)果 HRU組比HFD組的平均體質(zhì)量輕。HFD組睪丸和附睪組織中的甘油三酯的含量以及附睪組織中的甘油三酯的含量,明顯高于ND組和HRU組。相較于HFD組的睪丸和附睪均出現(xiàn)結(jié)構(gòu)松散,細(xì)胞結(jié)構(gòu)異常,管腔內(nèi)成熟的精子數(shù)量較少的現(xiàn)象,精子數(shù)量下降,活動度差等問題,HRU組均有明顯改善。解偶聯(lián)蛋白Ucp1基因和蛋白的表達(dá)量在蘆丁處理后顯著上升(P0.05),Mcp1和Tnfα的表達(dá)則顯著下降(P0.05)。結(jié)論蘆丁可以有效抑制高脂誘導(dǎo)的肥胖小鼠體質(zhì)量的快速增長,對肥胖誘發(fā)的生殖損傷有一定的保護(hù)效果。
[Abstract]:Objective to investigate the protective effect and mechanism of rutin on body mass inhibition and reproductive injury induced by obesity in obese male mice. Methods Twenty-four male ICR mice were randomly divided into 3 groups: normal control group (ND group, normal diet), high-fat diet group (HFD group, high-fat diet) and rutin protection group (HRU group, high-fat diet rutin group). According to the need of subsequent experiments, material selection is carried out. Biochemical indexes were detected by kit, paraffin sections of testis and epididymis were detected, morphological observation was performed by HE staining, sperm count, mobility and deformity rate were counted. The expression level of related genes was detected by fluorescence quantitative PCR.; Western blot was used to detect the expression of Ucp1 protein. Results the average body weight of HRU group was lighter than that of HFD group. The contents of triglyceride in testis and epididymis in HFD group and in epididymal tissue were significantly higher than those in ND group and HRU group. Compared with the HFD group, the testis and epididymis showed loose structure, abnormal cell structure, small number of mature sperm in the lumen, decreased sperm number and poor mobility. The expression of uncoupling protein Ucp1 gene and protein increased significantly after rutin treatment (P0.05), while the expression of Mcp1 and Tnf 偽 decreased significantly (P0.05). Conclusion rutin can effectively inhibit the rapid growth of body mass in obese mice induced by high fat, and has a protective effect on the reproductive injury induced by obesity.
