不同糖耐量人群血漿25-羥維生素D與葡萄糖轉(zhuǎn)運(yùn)子4及膽固醇調(diào)節(jié)元件結(jié)合蛋白1C表達(dá)關(guān)系的研究

【摘要】:目的探討不同糖耐量人群血漿25-羥維生素D[25(OH)D]與葡萄糖轉(zhuǎn)運(yùn)子4(GluT-4)及膽固醇調(diào)節(jié)元件結(jié)合蛋白1C(SREBP-1C)表達(dá)的關(guān)系。方法隨機(jī)分為T2DM組、IGR組及正常對照(NGT)組。檢測血糖、血脂、HbA1 c等,電化學(xué)發(fā)光法測定25(OH)D和FIns。Western blot檢測GluT-4及SREBP-1C蛋白的表達(dá),RT-PCR檢測GluT-4和SREBP-1C mRNA的表達(dá)。結(jié)果T2DM組、IGR組25(OH)D低于NGT組[(8.77±4.52)vs(18.35±5.90)vs(30.67±5.47)ng/ml,P0.05],T2DM組低于IGR組(P0.05)。T2DM組、IGR組血漿GluT-4 mRNA及蛋白表達(dá)低于NGT組[(0.235±0.412)vs(3.386±0.698)vs(6.325±0.959),(1.235±0.092)vs(2.430±0.153)vs(4.843±0.299),P0.05],T2DM組較IGR組降低(P0.05)。T2DM組、IGR組血漿中SREBP-1C mRNA及蛋白表達(dá)水平高于NGT組[(7.570±1.233)vs(5.561±0.820)vs(2.880±0.483),(8.340±1.092)vs(5.279±0.798)vs(2.340±0.135),P0.05],T2DM組較IGR組升高(P0.05)。Pearson相關(guān)性分析顯示,25(OH)D與血漿中GluT-4 mRNA、蛋白表達(dá)呈正相關(guān)(P0.05),與HbA_1 c、HOMA-IR、FIns、FPG、2 hPG、TG及SREBP-1C mRNA、SREBP-1C蛋白表達(dá)呈負(fù)相關(guān)(P0.05)。血漿中GluT-4表達(dá)與SREBP-1C表達(dá)呈負(fù)相關(guān)(P0.05)。結(jié)論血漿25(OH)D降低可引發(fā)糖耐量異常,甚至導(dǎo)致糖尿病,其機(jī)制可能與血漿中GluT-4表達(dá)降低及SREBP-1C表達(dá)升高有關(guān)。
[Abstract]:Objective to investigate the relationship between 25 hydroxyvitamin D [25 (OH) D] and the expression of glucose transporter 4 (GluT-4) and cholesterol regulatory element binding protein 1C (SREBP-1C) in patients with different glucose tolerance. Methods T2DM group, IGR group and normal control (NGT) group were randomly divided into three groups. Blood glucose, blood lipids and HbA1 c were detected. The expression of GluT-4 and SREBP-1C was detected by ECL and FIns.Western blot, and the expression of GluT-4 and SREBP-1C mRNA by RT-PCR. Results in T2DM group, 25 (OH) D in IGR group was lower than that in NGT group [(8.77 鹵4.52) vs (18.35 鹵5.90) vs (30.67 鹵5.47) ng/ml,P0.05], and that in T2DM group was lower than that in IGR group (P0.05). The expression of plasma GluT-4 mRNA and protein in IGR group was lower than that in NGT group [(0.235 鹵0.412) vs (3.386 鹵0.698) vs (6.325 鹵0.959), (1.235 鹵0.092) vs (2.430 鹵0.153) vs (4.843 鹵0.299), P0.05]. The expression of SREBP-1C mRNA and protein in T2DM group and IGR group was higher than that in NGT group [(7.570 鹵1.233) vs (5.561 鹵0.820) vs (2.880 鹵0.483)]. The protein expression was positively correlated (P0.05), and negatively correlated with the expression of hPG,TG and SREBP-1C mRNA,SREBP-1C protein in HbA_1. There was a negative correlation between GluT-4 expression and SREBP-1C expression in plasma (P0.05). Conclusion the decrease of plasma 25 (OH) D may lead to abnormal glucose tolerance and even lead to diabetes mellitus. The mechanism may be related to the decrease of GluT-4 expression and the increase of SREBP-1C expression in plasma.
