【摘要】：目的:通過(guò)檢測(cè)pSS患者血清中IL-27的水平和外周血白細(xì)胞表面IL-27R(gp130和WSX-1)的表達(dá)水平,分析IL-27及其受體與pSS發(fā)病的關(guān)系,探討IL-27及其受體在pSS發(fā)病中的意義。方法:(1)通過(guò)雙抗體夾心酶聯(lián)免疫吸附法來(lái)檢測(cè)pSS組與正常對(duì)照組外周血血清中IL-27的水平,分析pSS患者血清中IL-27的水平與pSS發(fā)病的關(guān)系。(2)通過(guò)流式細(xì)胞術(shù)來(lái)測(cè)定pSS組與正常對(duì)照組外周血白細(xì)胞表面的IL-27R(gp130和WSX-1)的表達(dá)水平,分析pSS患者白細(xì)胞表面IL-27R(gp130和WSX-1)的表達(dá)水平和pSS發(fā)病的關(guān)系。結(jié)果:(1)pSS組血清中IL-27的水平明顯高于正常對(duì)照組(161.25±22.75pg/ml VS 95.56±15.80pg/ml,p0.05);(2)pSS組外周血白細(xì)胞表面gp130(%)的表達(dá)水平(7.80±3.51)高于對(duì)照組(3.25±1.92)(p0.05),pSS組外周血白細(xì)胞表面WSX-1(%)的表達(dá)水平(2.91±0.99)高于對(duì)照組(1.45±0.85)(p0.05)。結(jié)論:pSS患者血清中的IL-27及白細(xì)胞表面的IL-27R(gp130和WSX-1)均存在異常表達(dá),提示IL-27及其受體可能參加了pSS的發(fā)生、發(fā)展過(guò)程。
[Abstract]:Objective: to analyze the relationship between IL-27 and its receptor and pSS by detecting the level of IL-27 in serum and the expression of IL-27R (gp130 and WSX-1) on peripheral blood leukocytes in patients with pSS, and to explore the significance of IL-27 and its receptor in the pathogenesis of pSS. Methods: (1) double antibody sandwich enzyme linked immunosorbent assay (Elisa) was used to detect the level of IL-27 in peripheral blood of pSS group and normal control group. To analyze the relationship between the level of IL-27 in serum of pSS patients and the pathogenesis of pSS. (2) to detect the expression of IL-27R (gp130 and WSX-1) on the surface of peripheral blood leukocytes in pSS group and normal control group by flow cytometry. To analyze the relationship between the expression of IL-27R (gp130 and WSX-1) and the pathogenesis of pSS in patients with pSS. Results: (1) the level of serum IL-27 in pSS group was significantly higher than that in normal control group (161.25 鹵22.75pg/ml VS 95.56 鹵15.80 PG / ml 0.05); (2). The expression of gp130 (%) on peripheral white blood cell surface in pSS group was (7.80 鹵3.51) higher than that in control group (3.25 鹵1.92) (p0.05), pSS group). The expression level of WSX-1 (%) on peripheral blood leukocyte surface in pSS group was (2.91 鹵0.99) higher than that in control group (1.45 鹵0.85) (p0.05). Conclusion: the abnormal expression of IL-27 in serum and IL-27R (gp130 and WSX-1) on leukocytes in patients with pSS suggests that IL-27 and its receptors may be involved in the pathogenesis and development of pSS.
