【摘要】：目的探究馬索羅酚(NDGA)對實驗性自身免疫性腦脊髓炎(EAE)小鼠血腦屏障的保護作用及其機制。方法將27只C57BL/6小鼠隨機分為正常組、EAE組與NDGA組,應用MOG35-55誘導EAE模型。小鼠發(fā)病后神經功能評分達1分時開始給予NDGA組NDGA 10 mg(/kg·d)腹腔注射,EAE組及正常對照組給予5%DMSO 10 ml/(kg·d)腹腔注射。給藥20 d后比較各組小鼠神經功能評分,應用熒光顯微鏡觀察伊文斯蘭染色后各組小鼠血腦屏障通透性改變情況,激光共聚焦顯微鏡觀察各組小鼠claudin-5、occludin、ZO-1表達情況,Real-time PCR檢測各組小鼠NF-κB及MMP-9 mRNA水平。結果與正常對照組相比,EAE組伊文斯蘭染色后熒光漏出范圍較大,緊密連接蛋白連續(xù)性遭到破壞,NF-κB、MMP-9 mRNA水平均上調;與EAE組相比,NDGA組神經功能評分顯著降低(P0.05),伊文斯蘭染色后熒光范圍較小且強度較弱,緊密連接蛋白連續(xù)性破壞減輕,NF-κB、MMP-9 mRNA水平下調(P0.05)。結論馬索羅酚可以保護EAE小鼠血腦屏障免遭破壞,延緩疾病進展,其機制可能是保護緊密連接蛋白完整性以及抑制MMP-9、NF-κB表達。
[Abstract]:Objective To investigate the protective effect of massolol (NDGA) on blood-brain barrier (BBB) in experimental autoimmune encephalomyelitis (EAE) mice and its mechanism.Methods Twenty-seven C57BL/6 mice were randomly divided into normal group, EAE group and NDGA group.The EAE model was induced by MOG35-55.The neurological function score of the mice was 1 minute after onset. Intraperitoneal injection, intraperitoneal injection of 5% DMSO 10 ml / (kg Results Compared with the normal control group, the fluorescence leakage range of EAE group was larger, the continuity of tight junction protein was destroyed, and the levels of NF-kappa B and MMP-9 mRNA were up-regulated in EAE group. Compared with EAE group, the neurological function score of NDGA group was significantly lower (P 0.05), and the fluorescence of Evans blue staining was up-regulated. Conclusion Masoprolol can protect the blood-brain barrier from damage and delay the progression of the disease in EAE mice. The mechanism may be to protect the integrity of tight junction protein and inhibit the expression of MMP-9 and NF-kappa B.
