脊髓小膠質(zhì)細(xì)胞酪氨酸家族激酶在糖尿病神經(jīng)痛中的作用

【摘要】:目的:探討脊髓背角小膠質(zhì)細(xì)胞上酪氨酸家族激酶(SFKs)在糖尿病神經(jīng)痛中的作用。方法:采用雄性SD大鼠,體重200~220 g,免疫熒光單染檢測(cè)腹腔注射鏈脲佐菌素(STZ)后7、14、28 d脊髓背角p-SFKs的表達(dá)水平,雙染觀察其表達(dá)部位;同時(shí)在給予STZ前鞘內(nèi)注射SFKs抑制劑PP2,行為學(xué)檢測(cè)大鼠50%機(jī)械撤足閾值的變化。結(jié)果 :與對(duì)照組比較,腹腔注射STZ后第1天,大鼠血糖水平開始升高(P0.001),持續(xù)至少28 d(P0.001);機(jī)械痛撤足逐漸下降(P0.05),28 d達(dá)到最低(P0.001);同時(shí)腹腔注射STZ后7、14、28 d,脊髓背角p-SFKs的表達(dá)水平明顯升高(均P0.01);只表達(dá)于小膠質(zhì)細(xì)胞中,神經(jīng)元和星型膠質(zhì)細(xì)胞中無(wú)表達(dá)。腹腔給予STZ前鞘內(nèi)注射PP2可防止STZ誘導(dǎo)的機(jī)械痛敏。結(jié)論:脊髓背角小膠質(zhì)細(xì)胞SFKs可能在糖尿病神經(jīng)痛中發(fā)揮重要作用。
[Abstract]:Aim: to investigate the role of tyrosine family kinase (SFKs) on spinal dorsal horn microglia in diabetic neuralgia. Methods: the expression of p-SFKs in spinal dorsal horn of male SD rats, weighing 200 ~ 220g, was detected by immunofluorescence single staining on the 28th day after intraperitoneal injection of streptozotocin (STZ). The expression sites of p-SFKs in spinal dorsal horn were observed by double staining. At the same time, the behavioral changes of 50% mechanical withdrawal threshold of rats were detected by intrathecal injection of SFKs inhibitor PP2, before STZ. Results: compared with the control group, the first day after intraperitoneal injection of STZ, The blood glucose level of rats began to rise (P0.001) and lasted for at least 28 days (P0.001); the mechanical pain gradually decreased (P0.05) and reached the lowest level at 28 days (P0.001); at the same time, the expression of p-SFKs in the dorsal horn of spinal cord increased significantly (all P0.01) at 714 days after intraperitoneal injection of STZ, and was only expressed in microglia cells. No expression was found in neurons and astrocytes. Intrathecal injection of PP2 before STZ could prevent mechanical pain induced by STZ. Conclusion: SFKs of spinal dorsal horn microglia may play an important role in diabetic neuralgia.
