【摘要】：目的研究肝X受體(LXRs)激動(dòng)劑T0901317對(duì)血糖波動(dòng)條件下人臍靜脈內(nèi)皮細(xì)胞(HUVEC)增殖能力的影響及其作用機(jī)制。方法用不同濃度的葡萄糖和T0901317處理HUVEC,采用CCK8法檢測(cè)HUVEC增殖情況;Western印跡法檢測(cè)Akt蛋白、p-Akt(Ser473)蛋白表達(dá)。結(jié)果血糖波動(dòng)抑制HUVEC的增殖及p-Akt(Ser473)蛋白的表達(dá)(P0.05)。T0901317促進(jìn)血糖波動(dòng)損傷后HUVEC的增殖(P0.05),上調(diào)p-Akt(Ser473)蛋白表達(dá)(P0.05),且具有濃度依賴性(P0.05)。此外,PI3K/Akt抑制劑LY294002(10μmol/L)可以阻斷T0901317上調(diào)p-Akt(Ser473)蛋白表達(dá)的作用(P0.05)。結(jié)論血糖波動(dòng)通過(guò)抑制PI3K/Akt信號(hào)通路減弱HUVEC增殖能力,LXRs激動(dòng)劑T0901317可通過(guò)激活PI3K/Akt信號(hào)通路增強(qiáng)血糖波動(dòng)抑制下HUVEC的增殖能力。
[Abstract]:Objective to study the effect of liver X receptor (LXRs) agonist T0901317 on the proliferation of human umbilical vein endothelial cells (HUVEC) and its mechanism. Methods HUVEC, with different concentrations of glucose and T0901317 were used to detect the proliferation of HUVEC by CCK8. Western blot was used to detect the expression of Akt protein p-Akt (Ser473). Results Blood glucose fluctuation inhibited the proliferation of HUVEC and the expression of p-Akt (Ser473) protein (P0.05). T0901317 promoted the proliferation of HUVEC (P0.05) and up-regulated the expression of p-Akt (Ser473) protein (P0.05) in a concentration-dependent manner (P0.05). In addition, PI3K / Akt inhibitor LY294002 (10 渭 mol/L) blocked the up-regulation of p-Akt (Ser473) protein expression by T0901317 (P0.05). Conclusion Blood glucose fluctuation attenuates the proliferation of HUVEC by inhibiting the PI3K/Akt signaling pathway. LXRs agonist T0901317 can enhance the proliferation of HUVEC by activating the PI3K/Akt signaling pathway.
