血紅素氧合酶系統(tǒng)對一氧化碳中毒后體外培養(yǎng)少突膠質(zhì)細(xì)胞Nogo-A的影響
[Abstract]:BACKGROUND: Nogo-A protein and its receptor NgR are expressed in oligodendrocytes and neurons of the central nervous system respectively, and the white matter demyelinates prominently in the brain imaging of patients with delayed encephalopathy induced by carbon monoxide poisoning. It is speculated that Nogo-A protein and NgR system may be involved in the brain damage induced by acute carbon monoxide poisoning and may be associated with delayed brain injury induced by carbon monoxide poisoning. Endogenous carbon monoxide (CO) is one of the most important gaseous messenger molecules in the body. It is mainly produced by the metabolism of heme oxygenase. Its effect on Nogo-A system has not been reported yet. Objective: To culture oligodendrocytes in vitro with exogenous CO treatment and inhibit the activity of heme oxygenase system with zinc protoporphyrin 9. Methods: Oligodendrocyte cultured in vitro were divided into control group, CO group and zinc protoporphyrin group 9. CO group and zinc protoporphyrin group 9 cells were cultured in sealed chamber containing 1% CO. Before CO treatment, zinc protoporphyrin group 9 cells were added to culture medium for 10 microns. Ol/L zinc protoporphyrin 9.Firstly, the expression levels of Nogo-A mR NA and protein in cultured cells at 6, 24 and 48 h were compared, and the differences of expression levels of Nogo-A mR NA and protein between CO and zinc protoporphyrin 9 cells at the highest expression levels were detected. The expression level of Nogo-A m RNA and protein in the nine zinc protoporphyrin groups was higher than that in the CO group at 24 h after culture. It can inhibit the expression level of Nogo-A m RNA and protein.
