硅油對(duì)淀粉樣蛋白和多肽積聚的促進(jìn)作用

【摘要】:淀粉樣疾病吸引了無(wú)數(shù)科學(xué)家的興趣,被Igor K. Lednev稱為世界第八大奇跡。其中,疏水界面對(duì)淀粉樣蛋白和多肽積聚的影響,自1994年起就陸續(xù)有文章報(bào)道。蛋白單體不僅富集在疏水界面上,并且在界面上采取有助于積聚的構(gòu)象和排列,甚至宏觀氣液界面也可顯著促進(jìn)α-突觸核蛋白(α-Syn)的積聚。 硅油是一類聚硅氧烷,呈疏水性,廣泛應(yīng)用于工業(yè)、醫(yī)療和日常用品中,通常被認(rèn)為是對(duì)人體安全無(wú)害的。例如,在眼科手術(shù)和美容整形中,常使用硅油作為填充物。水中的硅油微粒表面是一種疏水界面,但硅油對(duì)淀粉樣蛋白和多肽積聚的影響,尚未有報(bào)道。 本文采用硫磺素T(ThT)熒光法和原子力顯微鏡表征,研究了硅油對(duì)AcP11肽、α-突觸核蛋白(α-Syn)、淀粉樣蛋白β1-42(Human Aβ42)、酵母類Prion蛋白Ure2蛋白和GAV9肽的影響,,結(jié)果顯示,五種淀粉樣蛋白/多肽在含有硅油的體系中積聚更快、更充分,表明一定量的硅油能促進(jìn)這些淀粉樣蛋白/多肽的積聚。 此外,在常規(guī)的生物實(shí)驗(yàn)中,會(huì)使用離心管等容器,而且多數(shù)容器在制作時(shí),會(huì)對(duì)內(nèi)表面進(jìn)行硅油處理。本實(shí)驗(yàn)研究了15ml離心管在渦旋操作時(shí),對(duì)硅油污染的引入。結(jié)果表明,渦旋能將離心管內(nèi)壁的硅油引入溶液中,隨后促進(jìn)GAV9肽的積聚。 展望方面,調(diào)研了眼科手術(shù)中使用硅油填充后的并發(fā)癥,以白內(nèi)障較為突出,目前發(fā)病機(jī)制尚不清楚,在臨床歸納的可能因素中,并沒(méi)有提及硅油促進(jìn)淀粉樣蛋白積聚這一因素;而白內(nèi)障的發(fā)生,與晶狀體蛋白(Crystallin)的積聚密切相關(guān),可以推測(cè),填充后的白內(nèi)障并發(fā)癥,由硅油誘導(dǎo)促進(jìn)引起。
[Abstract]:Amyloid disease has attracted the interest of countless scientists, Igor K. Lednev called the world's eighth miracle. The effect of hydrophobic interface on the accumulation of amyloid and polypeptide has been reported since 1994. The protein monomers were not only concentrated at the hydrophobic interface, but also conformed and arranged in favor of accumulation at the interface. Even the macro-gas-liquid interface could significantly promote the accumulation of 偽 -synaptic nucleoprotein (偽 -Syn). Silicone oil is a kind of polysiloxane, which is hydrophobic and widely used in industrial, medical and daily necessities. For example, silicone oil is often used as a filler in eye surgery and cosmetic surgery. The surface of silicon oil particles in water is a hydrophobic interface, but the effect of silicone oil on amyloid and polypeptide accumulation has not been reported. The effects of silicone oil on AcP11 peptide, 偽 -syn, amyloid 尾 1-42 (Human A 尾 42, yeast Prion protein Ure2 protein and GAV9 peptide were studied by T (ThT) fluorescence and atomic force microscopy. The accumulation of five kinds of amyloid protein / polypeptide in the system containing silicone oil is faster and more sufficient, which indicates that a certain amount of silicone oil can promote the accumulation of these amyloid proteins / polypeptides. In addition, in conventional biological experiments, centrifuge tubes and other containers are used, and most containers are made with silicone oil on the inner surface. In this paper, the introduction of silicon oil pollution by 15ml centrifuge tube in vortex operation was studied. The results show that the vortex can introduce silicon oil from the inner wall of the centrifuge tube into the solution and then promote the accumulation of GAV9 peptide. In the aspect of prospect, the complications after silicone oil filling in ophthalmic surgery were investigated. Cataract is more prominent. At present, the pathogenesis of cataract is not clear, among the possible factors in clinical induction, However, the occurrence of cataract is closely related to the accumulation of crystallin (Crystallin). It can be inferred that the complications of cataract after filling are induced by silicone oil and promoted by silicone oil.
