新疆漢族2型糖尿病患者miR-375基因啟動(dòng)子區(qū)甲基化的初步研究

【摘要】:目的探討新疆漢族T2DM及糖耐量正常(NGT)人群miR-375啟動(dòng)子區(qū)部分CpG位點(diǎn)的甲基化狀態(tài)。方法采用基質(zhì)輔助激光解析電離時(shí)間飛行質(zhì)譜儀檢測100例T2DM患者(T2DM組)和100名NGT者(NGT組)外周血白細(xì)胞中miR-375基因啟動(dòng)子區(qū)5個(gè)CpG位點(diǎn)的甲基化狀態(tài),并對(duì)結(jié)果進(jìn)行分析。結(jié)果 (1)T2DM組平均甲基化水平高于NGT組(10.27%vs 7.24%,P0.01);(2)T2DM組miR-375基因5個(gè)CpG位點(diǎn)的甲基化率均高于NGT組,如CpG_22、CpG_24.25、CpG_26.27,僅CpG_26.27在兩組中比較,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論漢族T2DM患者外周血白細(xì)胞中miR-375基因部分啟動(dòng)子區(qū)異常甲基化水平可能與T2DM的發(fā)生有一定的關(guān)系。
[Abstract]:Objective to investigate the methylation status of partial CpG loci in miR-375 promoter region of T2DM and normal glucose tolerance (NGT) population in Xinjiang Han nationality. Methods the methylation status of 5 CpG sites in peripheral blood leukocytes of 100 T2DM patients (T2DM group) and 100 NGT subjects (NGT group) was detected by matrix assisted laser desorption time flight mass spectrometer. The results were analyzed. Results (1) the average methylation level of miR-375 gene in T2DM group was higher than that in NGT group (10.27%vs 7.24g / P0.01); (2). The methylation rate of miR-375 gene in T2DM group was higher than that in NGT group. Conclusion the abnormal methylation level of partial promoter region of miR-375 gene in peripheral blood leukocytes of Han T2DM patients may be related to the occurrence of T2DM.
