姜黃素對砷致大鼠睪丸生精細(xì)胞凋亡的拮抗作用

【摘要】:目的研究姜黃素對砷致大鼠睪丸生殖細(xì)胞凋亡的拮抗作用。方法將50只健康清潔級Wistar雄性大鼠隨機(jī)分為5組,分別為對照(雙蒸水)組和亞砷酸鈉(100 mg/L)染毒組及低、中、高劑量姜黃素(100 mg/L亞砷酸鈉+25、50、100mg/kg姜黃素)干預(yù)組,每組10只。亞砷酸鈉采用自由飲用方式進(jìn)行染毒;姜黃素采用經(jīng)口灌胃方式染毒,每周3次,連續(xù)8周。測量生精小管平均直徑;測定大鼠血清睪酮濃度及睪丸生精細(xì)胞凋亡指數(shù)。結(jié)果與對照組比較,亞砷酸鈉染毒組大鼠血清睪酮的水平降低,生精小管直徑縮小,而睪丸生精細(xì)胞凋亡指數(shù)升高,差異有統(tǒng)計學(xué)意義(P0.01)。與亞砷酸鈉染毒組比較,中、高劑量姜黃素干預(yù)組大鼠血清睪酮的水平升高,生精小管直徑增大,而睪丸生精細(xì)胞凋亡指數(shù)下降,差異均有統(tǒng)計學(xué)意義(P0.05)。且隨著姜黃素染毒劑量的升高,大鼠血清睪酮水平和生精小管直徑均呈上升趨勢,而睪丸生精細(xì)胞凋亡指數(shù)呈下降趨勢。結(jié)論姜黃素對砷致大鼠睪丸生殖細(xì)胞凋亡具有拮抗作用。
[Abstract]:Objective to study the antagonistic effect of curcumin on arsenic induced apoptosis of germ cells in rat testis. Methods Fifty healthy and clean grade Wistar male rats were randomly divided into five groups: control group (double distilled water), sodium arsenite (100 mg/L) and low dose curcumin (100 mg/L sodium arsenite 2550mg / kg curcumin) intervention group (10 rats in each group). Sodium arsenite was administrated by free drinking and curcumin was administrated by gavage three times a week for 8 weeks. The mean diameter of seminiferous tubules and the concentration of serum testosterone and the apoptosis index of testicular spermatogenic cells were measured. Results compared with the control group, the serum testosterone level and seminiferous tubule diameter were decreased and the apoptosis index of testicular spermatogenic cells was increased in the rats exposed to sodium arsenite (P0.01). Compared with sodium arsenite group, the serum testosterone level and seminiferous tubule diameter increased, while the apoptosis index of testicular spermatogenic cells decreased in high dose curcumin intervention group (P0.05). With the increase of curcumin dose, the serum testosterone level and seminiferous tubule diameter increased, while the apoptosis index of testicular spermatogenic cells decreased. Conclusion curcumin has an antagonistic effect on arsenic-induced apoptosis of germ cells in rat testis.
