金黃色葡萄球菌顯著增加系統(tǒng)性紅斑狼瘡患者中性粒細胞NETs產(chǎn)生

【摘要】:目的探討金黃色葡萄球菌對系統(tǒng)性紅斑狼瘡(SLE)中性粒細胞胞外誘捕網(wǎng)(NETs)產(chǎn)生的影響及意義。方法收集SLE穩(wěn)定者22例、活動者20例和健康對照18例,分離中性粒細胞并加金黃色葡萄球菌共培養(yǎng),檢測金黃色葡萄球菌刺激后NETs的主要成分,DNA試劑盒提取測定游離DNA(f DNA)及酶聯(lián)免疫吸附法測定(ELISA)上清LL-37,并分析其與SLE臨床及實驗指標的關(guān)系。結(jié)果 1)SLE穩(wěn)定組、活動組、健康對照組3組金黃色葡萄球菌刺激后f DNA量的比較差異有統(tǒng)計學意義,SLE活動組、穩(wěn)定組均高于健康對照組(P0.05),SLE活動組與穩(wěn)定組無顯著性差異(P0.05);LL-37比較無統(tǒng)計學意義(P0.05)。2)未經(jīng)金黃色葡萄球菌刺激f DNA量3組間差異有統(tǒng)計學意義,SLE穩(wěn)定組高于活動組和健康對照組(P分別是0.008、0.027),SLE活動組和健康對照組差異無統(tǒng)計學意義(P0.05);LL-37比較無統(tǒng)計學意義(P0.05)。3)3組體外金葡色葡萄球菌刺激后f DNA及LL-37量均較刺激前顯著增加,差異均有統(tǒng)計學意義(f DNA P分別是0.000 1、0.000 1、0.001,LL-37 P均為0.000 1);刺激前后f DNA的差值SLE活動組顯著高于穩(wěn)定組和健康對照組(P0.05),SLE穩(wěn)定組、健康對照組差異無統(tǒng)計學意義(P0.05);LL-37差值組間比較均無統(tǒng)計學意義(P0.05)。結(jié)論金黃色葡萄球菌刺激后,SLE患者NETs產(chǎn)生/釋放量顯著增加,SLE活動者最顯著,金黃色葡萄球菌對SLE疾病活動可能有影響。
[Abstract]:Objective to investigate the effect of Staphylococcus aureus on the production of neutrophil net (NETs) in systemic lupus erythematosus (SLE). Methods Neutrophils were isolated and co-cultured with Staphylococcus aureus in 22 stable SLE patients, 20 active SLE patients and 18 healthy controls. The free DNA (f DNA) and enzyme linked immunosorbent assay (Elisa) supernatant LL-37 were extracted from NETs stimulated by Staphylococcus aureus and the relationship between them and the clinical and experimental indexes of SLE was analyzed. Results 1) there were significant differences in the f DNA content after staphylococcus aureus stimulation among stable SLE group, active SLE group and healthy control group. There was no significant difference between the active SLE group and the stable group (P0.05) there was no significant difference between the stable group and the active group (P0.05). There was no significant difference between the stable SLE group and the active group (P0.05). There was no significant difference between the stable SLE group and the active group without staphylococcus aureus stimulation (P0.05). There was no significant difference between active SLE group and healthy control group (P < 0.05). There was no significant difference between LL-37 group and healthy control group (P0.05). After stimulation of staphylococcus aureus in vitro, the levels of f DNA and LL-37 were significantly increased in three groups. The differences were statistically significant (f DNA P was 0.000 ~ 0.000 1g ~ (-1) L ~ (L-37) P = 0.000 1), the difference of f DNA in active SLE group was significantly higher than that in stable SLE group and healthy control group (P 0.05), and the difference of f DNA in active SLE group was significantly higher than that in stable SLE group before and after stimulation (P0.05). There was no significant difference between the healthy control group (P0.05) and the difference of LL-37 was not statistically significant (P0.05). Conclusion the NETs production / release in patients with SLE after staphylococcus aureus stimulation is the most significant in SLE patients. Staphylococcus aureus may have an effect on disease activity of SLE.
