姜黃素對干熱環(huán)境下大鼠腎損傷保護(hù)作用的研究

【摘要】:目的:通過姜黃素預(yù)處理大鼠并復(fù)制干熱環(huán)境下大鼠中暑模型,探討姜黃素對干熱環(huán)境下大鼠腎損傷的保護(hù)作用及其可能機(jī)制。方法:成年SD雄性大鼠200只,隨機(jī)數(shù)字表法分為四:0分鐘組(常溫組)、50分鐘組、100分鐘組、150分鐘組。每組下分為五個(gè)亞組(n=10):鹽水(NS)組、溶劑(CMCNa)組、姜黃素50mg/kg組(Cur 50mg/kg)、姜黃素100mg/kg組(Cur 100mg/kg)、姜黃素200mg/kg組(Cur 200mg/kg)。鹽水組給予0.9%生理鹽水灌胃,溶劑組給予0.5%CMCNa溶液灌胃,姜黃素組大鼠給予不同濃度姜黃素溶液灌胃,連續(xù)七天,每日一次。復(fù)制干熱環(huán)境下大鼠中暑模型:將大鼠置于模擬沙漠干熱環(huán)境中(西北特殊環(huán)境人工實(shí)驗(yàn)艙,中國人民解放軍烏魯木齊總醫(yī)院研制),設(shè)置溫度41±0.5℃,濕度10±1%,每25分鐘監(jiān)測大鼠核心體溫。根據(jù)分組時(shí)間取出大鼠,進(jìn)行麻醉,腹主動(dòng)脈采血進(jìn)行生化檢測,穿刺膀胱留取尿液進(jìn)行腎損傷分子-1(KIM-1)及中性粒細(xì)胞明膠酶相關(guān)載脂蛋白(NGAL)的檢測,取腎組織用于病理及分子生物學(xué)檢測。結(jié)果:(1)在50min、100min及150min組,不同濃度的姜黃素處理后的大鼠血清中尿素、肌酐、KIM-1及NGAL的含量較同一時(shí)間點(diǎn)的鹽水(NS)組和溶劑組(CMCNa)明顯下降(P0.05)。(2)在干熱環(huán)境中,不同濃度的姜黃素可降低腎組織中MDA含量及提高SOD和CAT的活性,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。(3)在同一時(shí)間點(diǎn)干熱環(huán)境下,姜黃素預(yù)處理組大鼠腎組織內(nèi)IL-1β、IL-6和TNF-α的水平較鹽水(NS)組及溶劑(CMCNa)組降低,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。(4)在150min組,不同濃度的姜黃素預(yù)處理可明顯改善大鼠腎組織的病理改變。(5)大鼠腎組織石蠟切片TUNEL染色顯示,在150min組,姜黃素預(yù)處理可降低腎臟細(xì)胞的凋亡率,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。(6)在150min組,姜黃素預(yù)處理可降低腎組織內(nèi)炎癥反應(yīng)相關(guān)蛋白及凋亡相關(guān)蛋白的表達(dá)水平,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:(1)持續(xù)的干熱環(huán)境對大鼠腎臟具有明顯的損傷作用,并且隨著持續(xù)時(shí)間的延長而加重,腎組織內(nèi)炎癥反應(yīng)及氧化應(yīng)激水平升高,腎組織細(xì)胞凋亡顯著。(2)干熱環(huán)境下,不同濃度的姜黃素均可不同程度的減輕大鼠腎損傷,其中200mg/kg時(shí)效果最顯著。(3)姜黃素可能通過抑制炎癥及氧化應(yīng)激反應(yīng)、阻斷細(xì)胞凋亡通路,在一定程度上減輕了干熱環(huán)境下大鼠腎損傷。
[Abstract]:Aim: to investigate the protective effect of curcumin on renal injury and its possible mechanism by pretreating rats with curcumin and making heat stroke model in dry heat environment. Methods: 200 adult Sprague-Dawley male rats were randomly divided into 4: 0 minute group (normal temperature group), 50 minutes group, 100 minutes group and 150 minutes group. Each group was divided into five subgroups: saline group (NS), solvent group (CMCNa), curcumin 50mg/kg group (curcumin 50mg/kg), curcumin 100mg/kg group (curcumin 100mg/kg), curcumin 200mg/kg group (curcumin 200mg/kg). The saline group was given 0.9% normal saline, the solvent group was given 0.5 CMCNa solution, and the curcumin group was given curcumin solution of different concentrations once a day for seven days.
