維生素D對活動期系統(tǒng)性紅斑狼瘡患者內(nèi)皮祖細(xì)胞的影響

【摘要】:目的:觀察活動期系統(tǒng)性紅斑狼瘡(SLE)患者內(nèi)皮祖細(xì)胞(EPCs)數(shù)量及功能的變化,并探討維生素D對SLE患者EPCs數(shù)量及功能的影響.方法:收集30例SLEDAI評分大于8分,病情均處于活動期的SLE患者,30例年齡與性別匹配的健康人作為正常對照;酶聯(lián)免疫吸附試驗(ELISA)檢測外周血25羥基維生素D[25(OH)D]水平;密度梯度離心和貼壁培養(yǎng)法分離培養(yǎng)EPCs,流式細(xì)胞術(shù)檢測CD34~+/VEGFR-2~+EPCs在全血中的比例;通過計數(shù)再貼壁和構(gòu)建侵襲小室檢測EPCs黏附和遷移能力.結(jié)果:(1)體外培養(yǎng)過程中,活動期SLE患者EPCs數(shù)量(0.028±0.017)%顯著低于正常對照組(0.067±0.012)%,有統(tǒng)計學(xué)差異(P0.05);(2)活動期SLE患者EPCs遷移率(1.7±0.9)‰及黏附能力(19±7)顯著低于正常對照組EPCs遷移率(3.1±1.6)‰及黏附能力(34±11),有統(tǒng)計學(xué)差異(P0.05);(3)活動期SLE患者外周血25(OH)D質(zhì)量濃度(14.47±10.39)ng/mL水平低于健康對照組質(zhì)量濃度(24.15±7.98)ng/mL,有統(tǒng)計學(xué)差異(P0.05);(4)25(OH)D能夠增加活動期SLE患者EPCs數(shù)量(0.045±0.012)%,高于未加25(OH)D組(0.031±0.012)%,有統(tǒng)計學(xué)差異(P0.05);(5)25(OH)D能夠增加活動期SLE患者EPCs遷移率(2.6±0.7)‰及黏附能力(24±9),高于未加25(OH)D組遷移率(1.3±0.8)‰及黏附能力(13±6),有統(tǒng)計學(xué)差異(P0.05).結(jié)論:SLE患者維生素D水平降低,可誘發(fā)Ⅰ型干擾素通路活化,從而導(dǎo)致EPCs數(shù)量和/或功能異常,最終造成狼瘡患者血管內(nèi)皮損傷后修復(fù)障礙引發(fā)動脈粥樣硬化.
[Abstract]:Aim: to observe the changes of endothelial progenitor cells (EPCs) in active systemic lupus erythematosus (SLE) patients, and to investigate the effect of vitamin D on the number and function of EPCs in SLE patients. Methods: 30 SLE patients with SLEDAI score more than 8 and 30 healthy subjects matched with age and sex were collected as normal control, and the levels of 25 hydroxyvitamin D (25 (OH) D) in peripheral blood were detected by Elisa. EPCs were isolated and cultured by density gradient centrifugation and adherent culture. CD34 ~ / VEGFR-2 ~ EPCs were detected by flow cytometry, and the adhesion and migration of EPCs were detected by counting and reattachment and building invasive chambers. Results: (1) in vitro culture, The number of EPCs in active SLE patients (0.028 鹵0.017)% was significantly lower than that in normal controls (0.067 鹵0.012).
