地塞米松噴霧劑聯(lián)合牙髓干細(xì)胞治療糖尿病Beagle犬種植體周?chē)椎膶?shí)驗(yàn)研究

【摘要】:目的探索地塞米松噴霧劑聯(lián)合牙髓干細(xì)胞治療糖尿病(DM)Beagle犬種植體周?chē)讓?duì)骨吸收和炎癥因子的影響。方法 8只健康成年Beagle犬用四氧嘧啶誘導(dǎo)法構(gòu)建DM Beagle犬模型,分離培養(yǎng)牙髓干細(xì)胞,構(gòu)建DM Beagle犬種植體周?chē)啄P汀?只Beagle犬隨機(jī)分為對(duì)照組、實(shí)驗(yàn)A組、實(shí)驗(yàn)B組及實(shí)驗(yàn)C組,每組2只。對(duì)照組予以0.9%Na Cl每次2 m L,tid,噴霧+Bio-Oss骨粉+Bio-Gide膜覆蓋處理;實(shí)驗(yàn)A,B,C組在對(duì)照組治療的基礎(chǔ)上,分別將0.9%NaCl改為地塞米松噴霧劑、牙髓干細(xì)胞懸液、地塞米松噴霧劑+牙髓干細(xì)胞懸液,其余處理均相同。4組Beagle犬均治療24周。用X線片觀察術(shù)后第24周近、遠(yuǎn)中邊緣骨高度的變化,用酶聯(lián)免疫吸附(ELISA)法檢測(cè)齦溝液的腫瘤壞死因子-α(TNF-α)及白細(xì)胞介素-6(IL-6)的表達(dá)。結(jié)果術(shù)后第24周,對(duì)照組、實(shí)驗(yàn)A組、實(shí)驗(yàn)B組和實(shí)驗(yàn)C組的近中邊緣骨高度較術(shù)后的變化分別為(-0.42±0.14),(-0.36±0.12),(-0.29±0.11)和(-0.17±0.07)mm,遠(yuǎn)中邊緣骨高度較術(shù)后的變化分別為(-0.30±0.12),(-0.26±0.14),(-0.21±0.11)和(-0.12±0.09)mm,TNF-α分別為(82.72±19.31),(68.26±14.60),(54.78±14.43)和(42.51±12.95)ng·L~(-1),IL-6分別為(36.20±8.14),(31.32±9.36),(26.85±6.61)和(20.48±5.24)ng·L~(-1),實(shí)驗(yàn)C組的上述指標(biāo)和對(duì)照組比較,差異均有統(tǒng)計(jì)學(xué)意義(均P0.05)。結(jié)論地塞米松噴霧劑聯(lián)合牙髓干細(xì)胞治療可有效地降低DM Beagle犬種植體周?chē)椎难装Y因子水平及骨吸收。
[Abstract]:Objective to investigate the effects of dexamethasone spray combined with dental pulp stem cells on bone resorption and inflammatory factors in diabetic DMC Beagle dogs. Methods eight healthy adult Beagle dogs were induced by alloxan to construct DM Beagle canine model. Dental pulp stem cells were isolated and cultured, and the model of peri-implant inflammation of DM Beagle dog was established. Eight Beagle dogs were randomly divided into control group, experimental group A, experimental group B and experimental group C, with 2 dogs in each group. The control group was treated with 0.9%Na Cl 2 mL / time and sprayed with Bio-Gide membrane of Bio-Oss bone powder, and the experimental group of 0.9%NaCl was changed into dexamethasone spray and dental pulp stem cell suspension on the basis of the control group. Dexamethasone spray dental pulp stem cell suspension, other treatments were the same as 4 groups of Beagle dogs were treated for 24 weeks. The changes of bone height in the proximal and distal margin of gingival effusion were observed by X-ray film. The expression of TNF- 偽 (TNF- 偽) and interleukin-6 (IL-6) in gingival crevicular fluid were detected by enzyme linked immunosorbent assay (Elisa).
