miR-93及miR-21在糖尿病視網(wǎng)膜病變患者中的表達(dá)及其臨床價值

【摘要】:目的探討血漿微小核糖核酸(micro RNA,mi R)-93及mi R-21在糖尿病視網(wǎng)膜病變(diabetic retinopathy,DR)患者中的表達(dá)及其臨床價值。方法采用實(shí)時熒光定量PCR檢測98例DR患者、40例2型糖尿病(type 2 diabetes mellitus,DM)患者和40名健康體檢者(對照組)血漿中mi R-93及mi R-21的表達(dá)水平。應(yīng)用ROC曲線評價血漿mi R-93、mi R-21及兩項(xiàng)聯(lián)合對DR的診斷價值,多因素logistic回歸分析DR發(fā)生的危險因素。Pearson相關(guān)分析DR患者血漿mi R-93、mi R-21與各生化指標(biāo)的相關(guān)性。結(jié)果 DR組和DM組的尿白蛋白/肌酐、糖化血紅蛋白及空腹血糖均明顯高于對照組(均為P0.05),且DR組均明顯高于DM組(均為P0.05)。DR組血漿miR-93及miR-21表達(dá)水平(1.84±0.27、1.95±0.34)均明顯高于DM組(1.09±0.13、1.13±0.18)和對照組(1.02±0.08、1.08±0.16),差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。PDR組血漿miR-93及miR-21表達(dá)水平(2.31±0.57、2.28±0.61)均明顯高于NPDR組(1.40±0.21、1.52±0.24),差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。血漿miR-93、miR-21及兩項(xiàng)聯(lián)合診斷DR的曲線下面積(95%CI)分別為0.892(0.824~0.961)、0.836(0.770~0.905)、0.937(0.868~0.992)。尿白蛋白/肌酐、糖化血紅蛋白、空腹血糖、miR-93及miR-21是DR發(fā)生的的獨(dú)立危險因素。DR患者血漿miR-93、miR-21與尿白蛋白/肌酐、糖化血紅蛋白、空腹血糖均呈正相關(guān)(均為P0.01),且miR-93與miR-21呈正相關(guān)(P0.01)。結(jié)論血漿miR-93和miR-21與DR的發(fā)生發(fā)展密切相關(guān),可作為DR診斷的生物學(xué)標(biāo)志物。
[Abstract]:Objective to investigate the expression and clinical value of plasma microRNAi micro RNA-93 and miR-21 in patients with diabetic retinopathy. Methods the expression of miR-93 and miR-21 in plasma of 98 patients with Dr and 40 patients with type 2 diabetes mellitus (type 2 diabetes mellitus) and 40 healthy controls (control group) were detected by real-time fluorescence quantitative PCR. Results urinary albumin / creatinine was measured in Dr and DM groups. The levels of plasma miR-93 and miR-21 in Dr group were significantly higher than those in DM group (1.84 鹵0.27 鹵1.95 鹵0.34) and 1.09 鹵0.13 鹵0.18 in DM group and 1.02 鹵0.081.08 鹵0.16 in control group, respectively. The expression levels of miR-93 and miR-21 in P0.05).PDR group were significantly higher than those in NPDR group (2.31 鹵0.572.28 鹵0.61) and 1.40 鹵0.21 鹵1.52 鹵0.24 in NPDR group (all P 0.05). There was a positive correlation between fasting blood glucose (P 0.01) and miR-93 and miR-21 (P 0.01). Conclusion Plasma miR-93 and miR-21 are closely related to the occurrence and development of Dr, and can be used as biomarkers in the diagnosis of Dr.
