腺苷酸激活蛋白激酶與肥胖大鼠肝臟脂質(zhì)沉積的關(guān)系

【摘要】:目的探討肥胖大鼠肝臟組織腺苷酸激活蛋白激酶(AMPK)的表達及其對肝臟脂質(zhì)沉積的影響。方法選用18只剛斷乳的SPF級SD雄性大鼠,隨機挑選6只普通飼料喂養(yǎng)為正常組,另一組12只高脂飼料喂養(yǎng),12 w后成功建立肥胖模型大鼠6只。分別檢測體重、Lee指數(shù)、內(nèi)臟脂肪、肝濕重等指標。通過油紅O染色和檢測肝臟甘油三酯(TG)、總膽固醇(TC)含量評估兩組大鼠肝臟脂質(zhì)沉積的情況。采用Western印跡檢測大鼠肝臟磷酸化(p)-AMPK/AMPK、p-ACC/ACC水平。結(jié)果與正常組大鼠相比,肥胖組大鼠體重與Lee指數(shù)、附睪脂肪、腎周脂肪量、肝濕重、肝臟內(nèi)TG與TC含量均顯著升高,油紅O染色結(jié)果顯示肥胖組肝臟脂質(zhì)沉積較正常組明顯增多。Western印跡結(jié)果顯示,與正常組大鼠比較,肥胖組大鼠肝臟p-AMPK/AMPK和p-ACC/ACC表達明顯降低。兩組大鼠肝臟組織中p-AMPK、p-ACC表達水平與肝臟TG、TC呈負相關(guān)。結(jié)論高脂飲食誘導(dǎo)食源性肥胖大鼠可降低AMPK的活性,從而導(dǎo)致肝臟組織TG、TC合成增加,脂質(zhì)在肝臟組織中沉積增加。
[Abstract]:Objective to investigate the expression of adenylate activated protein kinase (AMPK) and its effect on lipids deposition in the liver of obese rats. Methods 18 newly weaned SPF grade SD male rats were randomly selected to be fed with normal diet (n = 6). Another group (n = 12) was fed with high fat diet for 12 weeks, and 6 obese rats were successfully established after feeding with high fat diet for 12 weeks. Lee index, visceral fat and wet weight of liver were measured. The lipids deposition in the liver of the two groups was evaluated by oil red O staining and the content of TGG (total cholesterol) in the liver. Western blot was used to detect the level of phosphorylated pAMPK / AMPKP p-ACC-ACC in rat liver. Results compared with the normal group, the body weight and Lee index, epididymal fat, perirenal fat, liver wet weight, TG and TC contents in the liver of obese rats were significantly increased. The results of oil red O staining showed that the lipid deposition in the obese group was significantly higher than that in the normal group. The results of Western blot showed that the expression of p-AMPK/AMPK and p-ACC/ACC in the obese group was significantly lower than that in the normal group. There was a negative correlation between the expression of p-AMPKT p-ACC and TGG TC in the liver of the two groups. Conclusion High fat diet can reduce the activity of AMPK in obese rats, which leads to the increase of TGT-synthesis and the increase of lipid deposition in liver tissue.
