【摘要】：目的觀察過(guò)表達(dá)硫酯酶超家族成員4(thioesterase superfamily member 4,THEM4)對(duì)高糖誘導(dǎo)的人腎小管上皮細(xì)胞膠原分泌的影響。方法體外培養(yǎng)人腎小管上皮細(xì)胞(human renal proximal tubular epithelial cell,HKC),將細(xì)胞隨機(jī)分為正常對(duì)照組(5.5 mmol·L~(-1)normal glucose,Control)、高糖組(30 mmol·L~(-1)high glucose,HG)、高糖+p Yrads-4-THEM4質(zhì)粒轉(zhuǎn)染組(HG+THEM4)、高糖+p Yr-adshuttle-4空質(zhì)粒對(duì)照組(HG+V)。后3組經(jīng)高糖刺激48 h后終止培養(yǎng),Western blot檢測(cè)THEM4、phospho-Akt(Ser473)、TGF-β1、α-SMA的表達(dá);免疫細(xì)胞熒光檢測(cè)THEM4蛋白表達(dá)和定位;ELISA方法檢測(cè)細(xì)胞上清液中Ⅰ型膠原(collagenⅠ,ColⅠ)、Ⅲ型膠原(collagenⅢ,ColⅢ)分泌情況。結(jié)果高糖刺激可導(dǎo)致人腎小管上皮細(xì)胞內(nèi)THEM4表達(dá)降低,phospho-Akt(Ser 473)、TGF-β1、α-SMA的表達(dá)增強(qiáng)及ColⅠ、ColⅢ分泌;過(guò)表達(dá)THEM4能夠逆轉(zhuǎn)高糖刺激所導(dǎo)致人腎小管上皮細(xì)胞phospho-Akt(Ser 473)蛋白的活化,下調(diào)α-SMA、TGF-β1表達(dá),抑制ColⅠ、ColⅢ的分泌。結(jié)論過(guò)表達(dá)THEM4能夠降低高糖誘導(dǎo)的膠原分泌,可能是通過(guò)抑制phospho-Akt(Ser 473)蛋白的活化,下調(diào)α-SMA、TGF-β1表達(dá)而實(shí)現(xiàn)的。
[Abstract]:Aim to investigate the effect of 4(thioesterase superfamily member 4 (THEM4), a member of the superfamily of thioesterase, on collagen secretion induced by high glucose in human renal tubular epithelial cells. Methods Human renal tubular epithelial cells were cultured in vitro.
