纈沙坦對糖尿病大鼠klotho蛋白及氧化應激影響的研究

【摘要】:目的探討纈沙坦對糖尿病大鼠腎臟klotho蛋白(KL)和氧化應激的影響。方法糖尿病大鼠隨機分為糖尿病(DM)組和纈沙坦干預(VA)組,另設對照(NC)組,每組20只。VA組予纈沙坦55mg/(kg·d)灌胃,NC組及DM組予等量生理鹽水灌胃,于4周及12周測定大鼠腎功能、血清KL、8-羥基脫氧鳥苷(8-OHdG)及超氧化物歧化酶(SOD)活性,免疫組織化學測定腎臟KL的表達水平。結果與NC組比較,DM組4周和12周時KL含量降低[(4.39±0.35)vs(3.73±0.69)ng/ml,(4.16±0.28)vs(3.30±0.37)ng/ml],SOD活性降低[(1.87±0.84)vs(1.69±0.29)U/ml,(1.79±0.48)vs(1.50±0.26)U/ml],8-OHdG增加[(2.74±0.11)vs(3.32±0.91)ng/ml,(2.86±0.28)vs(3.21±0.25)ng/ml](P0.05);與DM組比較,VA組上述指標亦改善,差異有統(tǒng)計學意義(P0.05)。結論糖尿病大鼠KL的表達降低,纈沙坦可提高糖尿病大鼠腎臟KL的表達,抑制氧化應激反應。
[Abstract]:Objective to investigate the effects of valsartan on renal klotho protein and oxidative stress in diabetic rats. Methods Diabetic rats were randomly divided into DM group and valsartan intervention group. Control group (n = 20) received valsartan 55mg/(kg d (n = 20) and DM group (n = 20) were treated with normal saline. Renal function was measured at 4 and 12 weeks.
