循環(huán)miRNA-93在DHEA多囊卵巢綜合征大鼠模型中的表達及其意義

【摘要】:目的:通過檢測多囊卵巢綜合征(PCOS)大鼠血清與卵巢組織中miRNA-93及血管內(nèi)皮生長因子A(VEGFA)的表達水平,探討循環(huán)miRNA-93及靶基因VEGFA在PCOS發(fā)生發(fā)展過程中的作用機理與診斷價值。方法:選取3~4周齡雌性(SD)大鼠,隨機分為實驗組和對照組,每組20只。實驗組每日頸背部皮下注射脫氫表雄酮(DHEA),按6mg/100g劑量溶于0.2ml無菌注射用油;對照組每日頸背部皮下注射0.2ml無菌注射用油劑。28天后觀察兩組大鼠的動情周期、卵巢形態(tài)學(xué)、性激素、體重、卵巢體積、OGTT血糖、空腹胰島素、胰島素抵抗指數(shù)(HOMA-IR)及血脂等代謝相關(guān)指標(biāo)的變化。RT-PCR法檢測大鼠血清和卵巢組織中miRNA-93表達,ELISA法檢測血清中VEGFA表達,IHC、Western blot法分析卵巢組織中VEGFA蛋白表達量。結(jié)果:實驗組大鼠失去規(guī)律的動情周期,卵巢體積增大,呈多囊樣變,血清FSH、T、LH升高,糖耐量異常,空腹胰島素及胰島素抵抗指數(shù)明顯高于對照組,血清膽固醇、甘油三酯高于對照組,PCOS大鼠模型構(gòu)建成功。RT-PCR、ELISA、Western blot、IHC等結(jié)果顯示,miRNA-93及靶基因VEGFA在PCOS大鼠模型的血清、卵巢組織中表達一致且顯著升高(P0.05)。結(jié)論:miRNA-93在PCOS大鼠模型血清和卵巢組織中呈高表達,可能通過調(diào)控其靶基因VEGFA在PCOS發(fā)生發(fā)展中起重要作用,在PCOS診斷中循環(huán)miRNA-93可能作為一種新型非侵入性的診斷標(biāo)記物。
[Abstract]:Objective: to investigate the mechanism and diagnostic value of circulating miRNA-93 and target gene VEGFA in the pathogenesis and development of polycystic ovary syndrome (PCOS) by detecting the expression of miRNA-93 and VEGF in serum and ovary of rats. Methods: three weeks old female SD rats were randomly divided into experimental group and control group with 20 rats in each group. The experimental group was injected with dehydroepiandrosterone (DHEAA) subcutaneously daily and dissolved in 0.2ml aseptic injection oil according to the dose of 6mg/100g. The oestrous cycle, ovary morphology and sex hormone were observed in the control group after subcutaneous injection of 0.2ml aseptic injection oil for the control group for 28 days. Body weight, ovarian volume, OGTT blood glucose, fasting insulin, Changes of insulin resistance index (HOMA-IRI) and serum lipids. RT-PCR was used to detect the expression of miRNA-93 in serum and ovarian tissues. Elisa was used to detect the expression of VEGFA in serum. Results: the rats in the experimental group lost the regular estrous cycle, increased the volume of ovary, presented polycystic degeneration, increased serum FSHT, LH, impaired glucose tolerance, and the fasting insulin and insulin resistance index were significantly higher than those in the control group. The results showed that the expression of miRNA-93 and target gene VEGFA in serum of PCOS rat model was consistent and significantly increased in ovarian tissue. Conclusion the high expression of 10 miRNA-93 in serum and ovarian tissue of PCOS rat model may play an important role in the pathogenesis and development of PCOS by regulating its target gene VEGFA. Circulating miRNA-93 may be a novel noninvasive diagnostic marker in the diagnosis of PCOS.
