胰島素樣生長因子1對糖尿病大鼠伴骨折模型骨折愈合的影響

【摘要】:目的探討胰島素生長因子(IGF)-1對糖尿病(DM)模型大鼠骨折后骨折愈合的影響及其機制。方法選取健康成年雄性SD大鼠40只,對所有大鼠進行編號后采用隨機數字表法選取10只作為正常組、其余30只大鼠采用腹腔注射鏈脲佐菌素建造糖尿病大鼠模型,建模成功后采用隨機數字表法分為IGF-1組、胰島素組、模型組各10只,各組大鼠均建立左脛骨上段骨折模型,分別給予對應的處理措施,分別于骨折后第1、2、4、6周觀察四組大鼠的骨折愈合情況、骨痂組織中相關指標的變化差異、血清骨鈣素等指標的含量差異。結果各組大鼠第1、2、4、6周血清骨鈣素含量差異顯著(P0.05),模型組顯著低于正常組、IGF-1組和胰島素組(P0.05),正常組、IGF-1組和胰島素組間比較無統(tǒng)計學差異(P0.05)。各組大鼠第1、2、4、6周血鈣、血清骨形態(tài)發(fā)生蛋白(BMP)-2、堿性磷酸酶(ALP)含量差異顯著(P0.05),模型組顯著低于正常組、IGF-1組和胰島素組(P0.05),在第1、2周時IGF-1組和胰島素組血鈣值、血清BMP-2、ALP水平明顯低于正常組(P0.05),在第4、6周無顯著差異(P0.05)。結論IGF-1對DM模型大鼠骨折具有促進骨折愈合的作用,其機制可能與提高血清骨鈣素、BMP-2、ALP及血鈣水平有關。
[Abstract]:Objective to investigate the effect and mechanism of insulin-growth factor (IGF-1) on fracture healing in rats with diabetes mellitus (DM). Methods A total of 40 healthy adult male SD rats were selected. 10 rats were selected as normal group by random number table method, and the other 30 rats were treated by intraperitoneal injection of streptozotocin (STZ) to construct diabetic rat model. After modeling successfully, the rats were randomly divided into IGF-1 group, insulin group and model group, 10 rats in each group. The model of left upper tibia fracture was established in each group, and the corresponding treatment measures were given respectively. The healing of fracture, the change of relative indexes in callus and the content of serum osteocalcin were observed at the 1st and 2nd week after fracture in the rats of the four groups. Results the level of serum osteocalcin was significantly lower in the model group than that in the normal group (P 0.05) and insulin group (P 0.05). There was no significant difference between the normal group and the insulin group (P 0.05). The levels of serum calcium, bone morphogenetic protein (BMPP) and alkaline phosphatase (ALP) were significantly lower in the model group than in the normal group (P 0.05). The serum calcium levels in the IGF-1 group and the insulin group were significantly lower than those in the normal control group and the insulin group at the 1st week. The level of serum BMP-2ALP was significantly lower than that of the normal group (P0.05A), and there was no significant difference in the level of BMP-2ALP at the 4th week (P0.05). Conclusion IGF-1 can promote fracture healing in DM model rats, and its mechanism may be related to the increase of serum osteocalcin, BMP-2, ALP and serum calcium levels.
