多囊卵巢綜合征患者顆粒細(xì)胞microRNA-200b的表達(dá)及影響

【摘要】:目的檢測(cè)多囊卵巢綜合征(PCOS)患者卵巢顆粒細(xì)胞中microRNA-200b(miR-200b)的表達(dá),分析其與胰島素抵抗的關(guān)系。方法選取PCOS患者120例(其中胰島素抵抗78例,非胰島素抵抗42例)及對(duì)照患者72例。采用逆轉(zhuǎn)錄定量PCR(qRT-PCR)檢測(cè)在卵巢顆粒細(xì)胞中miR-200b的表達(dá),并分析其臨床意義。結(jié)果 PCOS胰島素抵抗患者中超重或肥胖、高血壓、高血糖、脂代謝紊亂和代謝綜合征的發(fā)生率皆高于PCOS非胰島素抵抗患者和對(duì)照患者(P均0.05)。PCOS胰島素抵抗患者miR-200b表達(dá)上調(diào),與PCOS非胰島素抵抗患者和對(duì)照患者比較差異有統(tǒng)計(jì)學(xué)意義(P=0.012,P0.001);且PCOS胰島素抵抗患者中miR-200b表達(dá)量與胰島素抵抗指數(shù)(HOMA-IR)呈正相關(guān)(r=0.532,P=0.002)。結(jié)論 PCOS胰島素抵抗患者高血壓等代謝方面遠(yuǎn)期并發(fā)癥的患病率較高。miR-200b高表達(dá)與PCOS胰島素抵抗呈正相關(guān),可能是PCOS患者出現(xiàn)胰島素抵抗的重要病理機(jī)制之一。
[Abstract]:Objective to detect the expression of microRNA-200bmmiR-200b in granulosa cells of patients with polycystic ovary syndrome (PCOS) and to analyze the relationship between microRNA-200bmmiR-200b and insulin resistance.Methods 120 patients with PCOS (78 with insulin resistance and 42 with non-insulin resistance) and 72 controls were selected.The expression of miR-200b in ovarian granulosa cells was detected by reverse transcription quantitative PCR qRT-PCR and its clinical significance was analyzed.Results the incidences of overweight or obesity, hypertension, hyperglycemia, lipid metabolism disorder and metabolic syndrome in patients with PCOS insulin resistance were higher than those in patients with PCOS and control group. MiR-200b expression was up-regulated in patients with 0.05).PCOS insulin resistance.There was a significant difference between the PCOS patients with non-insulin resistance and the controls, and there was a positive correlation between the expression of miR-200b and the insulin resistance index (HOMA-IRI) in the patients with PCOS insulin resistance, and there was a positive correlation between the expression of miR-200b and the insulin resistance index (HOMA-IRI) in the patients with PCOS insulin resistance.Conclusion the high expression of .miR-200b in patients with PCOS insulin resistance is positively correlated with PCOS insulin resistance, which may be one of the important pathological mechanisms of insulin resistance in PCOS patients.
