【摘要】：目的·通過構建肥胖合并2型糖尿病(T2DM)Roux-en-Y胃旁路手術(RYGB)大鼠模型,探討RYGB對大鼠肝臟糖脂代謝的影響及其機制。方法·將大鼠隨機分為正常對照組、高脂飲食+DM組、假手術組、手術組,每組10只。監(jiān)測大鼠體質量、進食量、腹腔糖耐量試驗(IPGTT)以及胰島素耐量試驗(ITT)結果的變化,并檢測法尼醇X受體(FXR)、成纖維細胞生長因子21(FGF21)、葡萄糖-6-磷酸酶(G6Pase)的m RNA和蛋白表達水平。結果·與高脂飲食+DM組和假手術組比較,手術組大鼠體質量和進食量明顯下降,IPGTT、ITT曲線下面積明顯減小;肝臟油紅O染色可見手術組大鼠紅色脂滴較高脂飲食+DM組和假手術組明顯減少;手術組的FXR、FGF21 m RNA表達明顯升高,G6Pase m RNA表達降低;手術組FXR、FGF21蛋白表達明顯升高,G6Pase蛋白表達降低。結論·RYGB可以激活FXR-G6Pase通路,有效改善肥胖合并T2DM大鼠的糖脂代謝。
[Abstract]:Objective to investigate the effects of RYGB on hepatic glucose and lipid metabolism in rats with obesity and type 2 diabetes mellitus (T2DMX Roux-en-Y) by gastric bypass surgery. Methods Rats were randomly divided into normal control group, high fat diet DM group and sham operation group. The changes of body mass, food intake, intraperitoneal glucose tolerance test (IPGTT) and insulin tolerance test (ITT) were monitored in 10 rats in each group. The expression levels of m RNA and protein in FXRN, FGF21, G6 Pasein, FGF21 and G6 Pasein were determined, and the results were compared with those in DM group and sham operation group. The area under the ITT curve of IPGTT was significantly decreased in the operation group, and the red lipid droplets in the operation group were significantly lower than those in the DM group and the sham-operation group, and the liver oil red O staining showed that the red lipid droplets in the operation group were lower than those in the high fat diet DM group and sham-operation group. The expression of FXR- FGF21 m RNA was significantly increased in the operation group and decreased in the operation group, and the expression of FXR- FGF21 protein increased significantly in the operation group. Conclusion RYGB can activate the FXR-G6Pase pathway and effectively improve the glycolipid metabolism in the obese rats with T2DM.
