在有氧運(yùn)動(dòng)的基礎(chǔ)上服用補(bǔ)腎壯骨顆粒對(duì)改善去勢(shì)大鼠骨質(zhì)疏松癥病情的協(xié)同效果

【摘要】:目的探討有氧運(yùn)動(dòng)與補(bǔ)腎壯骨顆粒綜合療法治療絕經(jīng)后骨質(zhì)疏松癥(postmenopausal osteoporosis,PMOP)的療效及機(jī)理,為有效防治PMOP提供理論依據(jù)。方法采用去勢(shì)大鼠模型,分為假手術(shù)組(n=10)、模型組(n=10)、陽(yáng)性組(n=10)、有氧運(yùn)動(dòng)組(n=10)、補(bǔ)腎壯骨顆粒組(n=10)和聯(lián)合組(n=10),去勢(shì)后2 w運(yùn)用有氧運(yùn)動(dòng)、補(bǔ)腎壯骨顆粒以及二者聯(lián)合等干預(yù)方法防治大鼠發(fā)生的骨質(zhì)疏松。連續(xù)12 w檢測(cè)各組大鼠骨組織生物力學(xué),同時(shí)采用實(shí)時(shí)熒光定量PCR技術(shù)檢測(cè)骨代謝相關(guān)指標(biāo)表達(dá)。結(jié)果與假手術(shù)組相比,模型組大鼠的左股骨最大負(fù)荷、左脛骨最大負(fù)荷和第四腰椎結(jié)構(gòu)強(qiáng)度均顯著降低(P0.05)。與模型組相比,陽(yáng)性組、有氧運(yùn)動(dòng)組、補(bǔ)腎壯骨顆粒組和聯(lián)合組的左股骨最大負(fù)荷、左脛骨最大負(fù)荷和第四腰椎結(jié)構(gòu)強(qiáng)度均顯著提高(P0.05)。熒光定量PCR結(jié)果顯示,與模型組相比,陽(yáng)性組、有氧運(yùn)動(dòng)組、補(bǔ)腎壯骨顆粒組和聯(lián)合組的RANKL和OPG均顯著提高(P0.05),而RANKL/OPG顯著降低(P0.05)。與陽(yáng)性組相比,聯(lián)合干預(yù)組的RANKL和OPG均顯著提高(P0.05),而RANKL/OPG顯著降低(P0.05)。結(jié)論補(bǔ)腎壯骨顆粒和運(yùn)動(dòng)的綜合性治療策略能通過(guò)降低骨組織中OPG/RANKL比值,進(jìn)而抑制破骨細(xì)胞的活性來(lái)改善去勢(shì)大鼠骨質(zhì)疏松癥病情,其療效要優(yōu)于單純運(yùn)動(dòng)及補(bǔ)腎壯骨顆粒。
[Abstract]:Objective to explore the therapeutic effect and mechanism of aerobic exercise and Bushen Zhuanggu granule in treating postmenopausal osteoporosis (PMOP), and to provide theoretical basis for effective prevention and treatment of PMOP. Methods Ovariectomized rat model was used. They were divided into sham operation group, model group, positive group, aerobic exercise group, Bushen Zhuanggu granule group and combined group. Aerobic exercise was used 2 weeks after castration. Bushen Zhuanggu granule and its combination were used to prevent and treat osteoporosis in rats. Biomechanics of bone tissue in each group was detected for 12 weeks. At the same time, real-time fluorescence quantitative PCR was used to detect the expression of bone metabolism related indexes. Results compared with sham operation group, the maximal load of left femur in model group was higher than that in sham operation group. The maximum load of left tibia and the strength of the fourth lumbar vertebrae decreased significantly compared with the model group, the maximal load of left femur in positive group, aerobic exercise group, Bushen Zhuanggu granule group and combined group. The maximum load of left tibia and the structural strength of the fourth lumbar vertebra increased significantly. The results of fluorescence quantitative PCR showed that, compared with the model group, positive group, aerobic exercise group, Both RANKL and OPG in Bushen Zhuanggu granule group and combined group were significantly increased, while RANKL/OPG significantly decreased P0.05. In the combined intervention group, both RANKL and OPG increased significantly, while RANKL/OPG significantly decreased P0.05.Conclusion the comprehensive treatment strategy of Bushen Zhuanggu granule and exercise can reduce the ratio of OPG/RANKL in bone tissue. And then inhibit the activity of osteoclasts to improve the condition of osteoporosis in ovariectomized rats, its curative effect is better than simple exercise and Bushen Zhuanggu granule.
