【摘要】：本研究選擇分離自青海大骨節(jié)病區(qū)小麥麥穗中的一株優(yōu)勢內(nèi)生鏈格孢霉,探究其代謝產(chǎn)物對兔軟骨細(xì)胞的影響。通過顯微形態(tài)學(xué)觀察和ITS序列分析鑒定菌種;其代謝產(chǎn)物采用75%乙醇提取,然后依次用石油醚、乙酸乙酯、乙醚萃取;methylthiazolyldiphenyl-tetrazoliumbromide(MTT)法檢測兔軟骨細(xì)胞存活率;反轉(zhuǎn)錄-聚合酶鏈?zhǔn)椒磻?yīng)(reverse transcription and polymerase chain reaction)檢測軟骨細(xì)胞蛋白聚糖和Ⅱ型膠原蛋白的mRNA表達(dá)量;甲苯胺藍(lán)染色法(toluidine blue,TB)和免疫組化染色法(immunological histological chemistry,IHC)分別檢測蛋白聚糖和Ⅱ型膠原蛋白的表達(dá)。經(jīng)鑒定實(shí)驗(yàn)菌株(編號(hào)13-8-A)為互隔鏈孢霉(Alternaria alternate)。與對照組相比,實(shí)驗(yàn)組中的乙醇粗提物能抑制軟骨細(xì)胞生長,對蛋白聚糖和Ⅱ型膠原蛋白的表達(dá)及其mRNA的表達(dá)量均有抑制作用;其他萃取分部提取物都能不同程度地?fù)p傷軟骨細(xì)胞,但它們對這兩種蛋白的表達(dá)及其mRNA表達(dá)量的抑制均弱于相同濃度下乙醇粗提物的處理�；ジ翩滄呙�13-8-A代謝產(chǎn)物中應(yīng)該存在多種可以損傷軟骨細(xì)胞且可能具有協(xié)同作用的物質(zhì)。
[Abstract]:In this study, a dominant endophytic streptomyces was isolated from the wheat panicle of Kashin-Beck disease area in Qinghai Province, and the effect of its metabolites on rabbit chondrocytes was investigated. The strain was identified by microscopic observation and ITS sequence analysis. The metabolites were extracted by 75% ethanol, followed by petroleum ether, ethyl acetate and methyl thiazolyldiphenyl-tetrazoliumbromide MTT to determine the survival rate of rabbit chondrocytes.
