高脂飲食聯(lián)合鏈脲佐菌素對小鼠糖脂代謝及炎癥的影響

【摘要】:觀察高脂飲食聯(lián)合鏈脲佐菌素(STZ)對小鼠糖脂代謝及胰島功能損傷的影響。將體質(zhì)量為18~20 g的SPF級雄性C57BL/6J小鼠40只,隨機分為對照組和模型組,每組20只。對照組小鼠飼喂對照飼料,模型組小鼠飼喂高脂飼料。1周后,禁食16 h,測量小鼠體質(zhì)量與空腹血糖;尾靜脈采血,分離血清;每周1次,連續(xù)4周。4周后,對照組小鼠腹腔注射檸檬酸緩沖液,模型組小鼠腹腔注射STZ,劑量為40 mg·kg~(-1),每天1次,連續(xù)3 d。繼續(xù)飼養(yǎng)2周后,測量隨機血糖,以小鼠血糖≥16.7 mmol·L~(-1)者即判定為2型糖尿病。對照組及2型糖尿病小鼠經(jīng)腹腔注射葡萄糖以進行糖耐量試驗。與對照組同一時間的體質(zhì)量和血糖進行比較,模型組小鼠體質(zhì)量、血糖均升高,除第1周血糖外,差別均有統(tǒng)計學(xué)意義(P0.05)。采用重復(fù)測量方差分析,發(fā)現(xiàn)喂養(yǎng)時間對體質(zhì)量(F=200.831)和血糖(F=7.025)均有影響,差異有統(tǒng)計學(xué)意義(P0.05);不同喂養(yǎng)時間對低密度脂蛋白(F=30.793)、甘油三酯(F=34.027)和高密度脂蛋白(F=30.793)均有影響,差異有統(tǒng)計學(xué)意義(P0.05)。不同飼料喂養(yǎng)與不同喂養(yǎng)時間對體質(zhì)量和甘油三酯存在交互作用(P0.05)。比較糖耐量曲線發(fā)現(xiàn),成模小鼠較對照組小鼠糖耐量降低。采用ELISA試劑盒檢測小鼠血清中TNF-α含量,成模小鼠的TNF-α含量高于對照組,差異有統(tǒng)計學(xué)意義(P0.05)。高脂飲食可導(dǎo)致C57BL/6J小鼠糖脂代謝紊亂,給予STZ后引起了小鼠糖耐量降低及炎癥損傷,高脂飲食聯(lián)合STZ可提高小鼠2型糖尿病模型的成模率。
