建立雄性大鼠骨質(zhì)疏松模型的方法學(xué)研究

【摘要】:目的探討藥物誘導(dǎo)與手術(shù)去勢構(gòu)建雄性大鼠骨質(zhì)疏松模型的可行性,為進(jìn)一步研究男性骨質(zhì)疏松提供支持。方法雄性SD大鼠40只,隨機(jī)分為4組(每組10只):空白對照組、手術(shù)組、給藥組和共處理組。對照組未做處理;手術(shù)組行雙側(cè)睪丸切除方法;給藥組行皮下注射D-半乳糖;共處理組同時行手術(shù)和皮下注射D-半乳糖。于干預(yù)前及干預(yù)后第8周測定體重,骨密度和骨礦鹽含量,組織、脂肪、肌肉含量。結(jié)果術(shù)后第8周,三組實(shí)驗(yàn)組骨密度較對照組骨密度降低,共處理組和手術(shù)組骨密度分別下降12%和7%,統(tǒng)計(jì)學(xué)差異顯著(P0.01);給藥組骨密度下降2%,不存在統(tǒng)計(jì)學(xué)意義(P0.05);各組骨礦鹽含量無差異(P0.05)。共處理組骨密度較手術(shù)組骨密度低5.6%,肌肉含量低19.8%,存在統(tǒng)計(jì)學(xué)差異(P0.05)。四組間體重?zé)o統(tǒng)計(jì)學(xué)差異(P0.05);三組實(shí)驗(yàn)組均低于對照組脂肪含量,統(tǒng)計(jì)學(xué)差異顯著(P0.01),三組間脂肪含量無統(tǒng)計(jì)學(xué)差異(P0.05)。結(jié)論本研究手術(shù)去勢法8周可以建立骨質(zhì)疏松模型;去勢法和皮下注射D-半乳糖聯(lián)合使用8周建立骨質(zhì)疏松模型骨密度下降更為明顯;雄性動物骨質(zhì)疏松后,脂肪含量減少。
[Abstract]:Objective to explore the feasibility of establishing osteoporosis model of male rats by drug induction and castration, and to provide support for further study on osteoporosis. Methods Forty male SD rats were randomly divided into 4 groups (10 rats in each group: blank control group). Operation group, administration group and co-treatment group. The control group was not treated, the operation group received bilateral orchiectomy, the drug group received subcutaneous injection of D-galactose, and the control group received subcutaneous injection of D-galactose. The body weight, bone mineral density, bone mineral salt content, tissue, fat and muscle content were measured before and 8 weeks after intervention. The BMD of the three experimental groups was lower than that of the control group. The BMD of co-treatment group and operation group decreased by 12% and 7, respectively, the difference was significant (P0.01), the BMD of administration group was decreased by 2%, there was no significant difference in bone mineral salt content in each group (P 0.05). The BMD of co-treatment group was lower than that of operation group (P < 0.05). 5.6The muscle content was 19.8%, there was statistical difference between the four groups (P 0.05), there was no significant difference in body weight among the four groups (P 0.05), and the fat content in the three groups was lower than that in the control group. There was no significant difference in fat content among the three groups (P 0.05). Conclusion Osteoporosis model can be established by ovariectomy for 8 weeks. The bone mineral density of 8 Zhou Jianli osteoporosis model decreased more obviously by castration and subcutaneous injection of D-galactose, and the fat content decreased after osteoporosis in male animals.
