骨髓間充質(zhì)干細(xì)胞移植對睪丸間質(zhì)細(xì)胞再生及睪酮分泌的影響

【摘要】:背景:干細(xì)胞移植治療雄性激素部分性缺乏綜合征鮮有報(bào)道。目的:觀察骨髓間充質(zhì)干細(xì)胞移植對老年大鼠性激素水平及睪丸組織的影響。方法:取30只6月齡SD大鼠作為正常對照組,檢測血清睪酮和游離睪酮濃度;選擇血清睪酮和游離睪酮濃度低于正常對照組的90只20月齡SD大鼠,作為雄性激素部分性缺乏綜合征模型,隨機(jī)分為3組,模型組、原位移植組與尾靜脈移植組,原位移植組在睪丸內(nèi)注射2×10~8 L~(-1)的骨髓間充質(zhì)干細(xì)胞懸液0.2 m L,尾靜脈移植組經(jīng)尾靜脈注射2×10~8 L~(-1)的骨髓間充質(zhì)干細(xì)胞懸液0.5 m L。移植8周后,檢測各組血清中卵泡刺激素、促黃體激素及睪酮水平,并制作睪丸組織病理切片,觀察睪丸組織及間質(zhì)細(xì)胞情況。結(jié)果與結(jié)論:(1)血清檢測結(jié)果:4組間卵泡刺激素、促黃體激素水平比較差異均無顯著性意義;模型組睪酮水平明顯低于正常對照組(P≤0.01),原位移植組、尾靜脈移植組睪酮水平明顯高于模型組(P≤0.01);(2)蘇木精-伊紅染色結(jié)果:正常對照組睪丸組織完整,可見大量間質(zhì)細(xì)胞、支持細(xì)胞及各級生精細(xì)胞;模型組睪丸組織缺失,支持細(xì)胞與間質(zhì)細(xì)胞稀疏,缺乏精原細(xì)胞與生精細(xì)胞;原位移植組、尾靜脈移植組睪丸組織較完整,可見大量間質(zhì)細(xì)胞與支持細(xì)胞或生精細(xì)胞;(3)NBT染色結(jié)果:模型組睪丸間質(zhì)細(xì)胞數(shù)量明顯少于正常對照組(P≤0.01),原位移植組、尾靜脈移植組睪丸間質(zhì)細(xì)胞數(shù)量明顯多于模型組(P≤0.01);(4)結(jié)果表明:骨髓間充質(zhì)干細(xì)胞移植對老年大鼠睪丸組織有修復(fù)作用。
[Abstract]:Background: stem cell transplantation in the treatment of male hormone partial deficiency syndrome is rarely reported. Objective: to observe the effect of bone marrow mesenchymal stem cell transplantation on sex hormone level and testicular tissue in aged rats. SD rats served as normal control group, Serum testosterone and free testosterone levels were measured, 90 SD rats aged 20 months with serum testosterone and free testosterone concentrations lower than normal control group were selected as male hormone partial deficiency syndrome model, and were randomly divided into 3 groups, model group. In the orthotopic transplantation group and the tail vein transplantation group, the bone marrow mesenchymal stem cell suspension (0.2 mL) was injected into the testis of the orthotopic transplantation group (2 脳 10 8 L), and the bone marrow mesenchymal stem cell suspension (0.5 mL) was injected through the tail vein through the tail vein transplantation group.
