【摘要】:類風(fēng)濕關(guān)節(jié)炎(Rheumatoid arthritis,RA)病人滑膜成纖維細(xì)胞(Fibroblast-like synoviocytes,FLS)是研究RA發(fā)病機(jī)制和進(jìn)行RA藥物篩選的最理想的細(xì)胞來源.但是對(duì)于眾多非醫(yī)療機(jī)構(gòu)的高校和科研院所而言,由于RA病人滑膜組織不易獲取而難以進(jìn)行細(xì)胞水平的操作.因此,建立膠原誘導(dǎo)的關(guān)節(jié)炎大鼠(Collagen-induced arthritis,CIA)模型,成功分離膝關(guān)節(jié)滑膜組織并進(jìn)行滑膜成纖維細(xì)胞的原代培養(yǎng)和純化,將有效解決RA FLS的來源問題,為后續(xù)實(shí)驗(yàn)奠定基礎(chǔ).本實(shí)驗(yàn)通過皮內(nèi)和皮下兩次注射雞Ⅱ型膠原誘導(dǎo)Wistar大鼠罹患關(guān)節(jié)炎,通過關(guān)節(jié)炎指數(shù)評(píng)分和膝關(guān)節(jié)HE染色明確CIA模型的建立;分離膝關(guān)節(jié)滑膜組織,采用Ⅱ型膠原酶和無EDTA胰酶消化法分離細(xì)胞并進(jìn)行傳代培養(yǎng),傳至第三代時(shí),細(xì)胞免疫熒光檢測(cè)細(xì)胞中VCAM-1蛋白含量以鑒定細(xì)胞純度.結(jié)果顯示,滑膜組織經(jīng)過消化和培養(yǎng)后得到大量大鼠滑膜成纖維細(xì)胞;在顯微鏡下觀察所得細(xì)胞呈梭形樹突狀,具有成纖維樣細(xì)胞特征;VCAM-1蛋白陽性細(xì)胞達(dá)98%以上,確證所分離細(xì)胞為滑膜成纖維樣細(xì)胞,且純度極高.本研究建立了較簡(jiǎn)便快捷地獲得大量滑膜成纖維細(xì)胞的方法,為RA的發(fā)病機(jī)制研究和藥物防治提供了豐富的實(shí)驗(yàn)材料.
[Abstract]:Fibroblast-like synoviocytes-FLSs (Fibroblast-like synoviocytes-FLSs) from patients with rheumatoid arthritis (Rheumatoid arthritis RAA) are ideal cell sources for studying the pathogenesis of RA and screening RA drugs. But for many non-medical institutions and research institutes, Because synovial tissue of RA patients is difficult to obtain, it is difficult to operate at cell level. Therefore, Collagen-induced arthritis CIAA model of collagen-induced arthritis rat was established, and synovial tissue of knee joint was isolated successfully and primary culture and purification of synovial fibroblasts were carried out.
