肥胖對(duì)胰島β細(xì)胞凋亡的影響

【摘要】:目的探討肥胖對(duì)胰島β細(xì)胞凋亡的影響及其可能的機(jī)制。方法取雄性SD大鼠,采用高脂飲食喂養(yǎng)的方法建立大鼠肥胖模型。取肥胖模型組(obese model group,OM組)和正常對(duì)照組(normal control group,NC組)大鼠各10只檢測(cè)。測(cè)定空腹血糖(FBG)、總膽固醇(TC)、三酰甘油(TG)、游離脂肪酸(FFA)及血清胰島素(FIns)的水平,計(jì)算胰島素抵抗指數(shù)(HOMA-IR)。蘇木精-伊紅染色觀察胰腺組織病理變化;脫氧核糖核苷酸末端轉(zhuǎn)移酶介導(dǎo)的缺口末端標(biāo)記法(TUNEL)檢測(cè)β細(xì)胞凋亡水平;Western blot檢測(cè)細(xì)胞色素C(Cyt C)在線粒體、細(xì)胞質(zhì)中的分布情況及活化的胱天蛋白酶-3(Caspase-3)的表達(dá)水平。結(jié)果與NC組比較,OM組大鼠FBG、TC、TG、FFA、FIns、HOMA-IR、凋亡指數(shù)、胞質(zhì)Cyt C水平及活化的Caspase-3表達(dá)水平均明顯升高;線粒體Cyt C水平明顯降低(均P0.05);胰腺組織出現(xiàn)結(jié)構(gòu)破壞、脂質(zhì)變性等改變。結(jié)論肥胖通過線粒體途徑增加胰島β細(xì)胞的凋亡水平。
[Abstract]:Objective to investigate the effect of obesity on islet 尾 cell apoptosis and its possible mechanism. The obesity model of rats was established by high fat diet. The obesity model group (10 rats) and the normal control group group (NC group) were used to detect the fasting blood glucose (FBG), total cholesterol (TC), triacylglycerol (TG) and free lipid (FG) in the obese model group (n = 10) and the normal control group (n = 10). The levels of fatty acid FFAand serum insulin FIns, The insulin resistance index (HOMA-IRN), hematoxylin-eosin staining (HE staining), deoxyribonucleotidyl transferase-mediated Nick end labeling (DNT-dTTP) -mediated dUTP Nick end labeling (Tunel), and cytochrome C (Cyt C) in mitochondria were measured by blot, and the levels of apoptosis of 尾 cells were detected by Tunel Nick end labeling (Tunel). Results compared with the NC group, the expression of Caspase-3 in the cytoplasm and the expression of activated cystatin 3 were significantly higher in the OM group than in the control group. The apoptotic index, the level of cytoplasmic Cyt C and the expression of activated Caspase-3 were significantly higher in the OM group than in the control group. The level of mitochondrial Cyt C was significantly decreased (all P0.05%; pancreatic tissue structure was damaged, lipid denaturation and other changes). Conclusion Obesity can increase the apoptosis level of islet 尾 cells through mitochondrial pathway.
