【摘要】：目的研究血清素與SD大鼠成骨細(xì)胞(Osteoblasts,OBs)的劑量相關(guān)性。方法(1)貼壁培養(yǎng)與分離新生大鼠顱骨細(xì)胞。通過(guò)觀察細(xì)胞形態(tài)和熒光染色鑒定。(2)向P3代細(xì)胞培養(yǎng)液中混進(jìn)不同濃度(10-5 mol/L、10-6 mol/L、10-7 mol/L、10-8 mol/L、10-9 mol/L、0mol/L)血清素干預(yù)。(3)第5天用MTT法分析干預(yù)因素與增殖的關(guān)系。(4)第10天分別用微量酶標(biāo)法測(cè)堿性磷酸酶(Alkaline phosphatase,ALP)水平和免疫熒光法測(cè)Ⅰ型膠原蛋白(Collagen-Ⅰ,Col-Ⅰ)水平分析干預(yù)因素與分化的關(guān)系。(5)第15天用ELISA法測(cè)骨鈣素(Osteocalcin,OCN)水平,分析干預(yù)因素與分化的關(guān)系。統(tǒng)計(jì)學(xué)方法通過(guò)SPSS 20.0收集整理并分析數(shù)據(jù)。計(jì)量資料用均數(shù)±標(biāo)準(zhǔn)差((?)±s)表示,不同組間比較采用單因素方差分析,多組間兩兩之間比較采用LSD-t檢驗(yàn),P0.05表示差異有統(tǒng)計(jì)學(xué)意義。結(jié)果不同濃度(10-5 mol/L、10-6 mol/L、10-7 mol/L、10-8 mol/L、10-9 mol/L、0mol/L)血清素干預(yù),MTT法測(cè)得平均光密度值分別為:0.400±0.028,0.379±0.023,0.379±0.047,0.401±0.038,0.420±0.038,0.618±0.064,任意組與空白對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05);微量酶標(biāo)法測(cè)得ALP平均水平分別為:0.046±0.002,0.017±0.002,0.026±0.002,0.046±0.002,0.051±0.002,0.073±0.003,任意組與空白對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05);免疫熒光法測(cè)得Col-Ⅰ平均水平分別為:45.581±6.378,33.110±1.161,23.327±0.784,15.731±2.243,27.331±2.430,33.592±2.684,除10-6 mol/L組外任意組與空白對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05);ELISA法測(cè)得OCN平均水平分別為:3.002±1.639,1.711±0.370,2.193±0.551,2.439±1.469,2.703±1.175,8.021±3.036,任意組與空白對(duì)照組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論血清素對(duì)成骨細(xì)胞成負(fù)性效應(yīng),這種負(fù)效應(yīng)與血清素濃度呈劑量相關(guān)性,隨其濃度增高呈先強(qiáng)后弱關(guān)系。
[Abstract]:Objective to study the dose correlation between serotonin and osteoblasts of SD rats.
