膳食鈣對(duì)飲水型氟暴露致子代肝臟損傷的干預(yù)作用

【摘要】:目的探討膳食鈣對(duì)氟暴露致子代肝臟損傷的分子機(jī)制。方法取初斷乳SD大鼠雌鼠100只,雄鼠50只。雌鼠隨機(jī)分成對(duì)照、染氟、低鈣、低鈣染氟、高鈣染氟5組。3個(gè)月后合籠,取14和28日齡仔鼠進(jìn)行相關(guān)實(shí)驗(yàn)。結(jié)果與對(duì)照組相比,其他各組仔鼠肝細(xì)胞的凋亡數(shù)量明顯增多,28日齡仔鼠低鈣染氟組Bcl-2蛋白表達(dá)降低(P0.05),染氟組和低鈣染氟組14日齡雄性、28日齡雌雄仔鼠肝組織Caspase12蛋白表達(dá)水平升高(P0.01);與染氟組相比,低鈣染氟組的細(xì)胞凋亡數(shù)量明顯增多,高鈣染氟組的細(xì)胞凋亡數(shù)量有所下降,各日齡仔鼠低鈣染氟組Bcl-2蛋白表達(dá)降低(P0.05),28日齡雄性仔鼠高鈣染氟組Caspase12蛋白表達(dá)降低(P0.05),低鈣組降低(P0.01)。結(jié)論氟中毒致子代肝損傷的機(jī)制可能是氟暴露導(dǎo)致子代肝組織上調(diào)內(nèi)質(zhì)網(wǎng)途徑凋亡通路中Caspase12表達(dá),下調(diào)Bcl-2的表達(dá),低鈣與染氟表現(xiàn)出協(xié)同作用,高鈣則能緩解氟中毒癥狀。
[Abstract]:Objective to investigate the molecular mechanism of liver injury induced by dietary calcium exposure to fluoride. Methods 100 female SD rats and 50 male rats were randomly divided into control group, low calcium group, low calcium fluoride group and high calcium fluoride group. The 14 and 28 day old rats were selected to carry out the related experiments. The results were compared with those of the control group. The number of hepatocytes apoptosis in other groups increased significantly. The expression of Bcl-2 protein decreased in the low calcium fluoride group of 28 days old rats, while the expression level of Caspase12 protein in the liver tissue of the 14 day old male male rats exposed to fluorine and the low calcium fluoride group increased significantly (P 0.01), compared with the fluoride group, the expression of Caspase12 protein in the liver tissue of the rats exposed to fluoride was significantly higher than that of the control group. The number of apoptosis increased significantly in low calcium fluoride group, but decreased in high calcium fluoride group. The expression of Bcl-2 protein decreased in the low calcium fluoride group and the low calcium group in 28 days old male rats. Conclusion the mechanism of liver injury induced by fluorosis may be fluorine exposure induced by fluorosis in offspring liver tissue. Upregulate the expression of Caspase12 in the endoplasmic reticulum pathway. The expression of Bcl-2 was down-regulated. Low calcium and fluoride showed synergistic effect, while high calcium could relieve the symptoms of fluorosis.
