【摘要】：目的探討高糖高溶血磷脂膽堿環(huán)境對(duì)人腎小球系膜細(xì)胞(HMCs)產(chǎn)生纖維連接蛋白(Fn)、Ⅳ型膠原(ColⅣ)及轉(zhuǎn)化生長(zhǎng)因子-β1(TGF-β1)的影響。方法將HMCs分為對(duì)照組、TGF-β1中和性抗體組、高糖高脂組、高糖高脂+TGF-β1中和性抗體組,采用qRT-PCR檢測(cè)TGF-β1mRNA的表達(dá),ELISA檢測(cè)細(xì)胞上清液中ColⅣ及Fn含量。結(jié)果 (1)高糖高脂組TGF-β1mRNA表達(dá)水平較對(duì)照組升高(P0.05);(2)高糖高脂組細(xì)胞上清液TGF-β1、ColⅣ和Fn含量較對(duì)照組增加(P0.05),而這種作用可被TGF-β1中和性抗體所抑制(P0.05)。結(jié)論高糖高脂可通過(guò)促進(jìn)系膜細(xì)胞TGF-β1表達(dá)從而誘導(dǎo)ECM的分泌,增加腎纖維化發(fā)生的風(fēng)險(xiǎn)。
[Abstract]:Objective to investigate the effects of high glucose and high lysophosphatidylcholine (LPC) environment on the production of fibronectin (FN) and transforming growth factor- 尾 1 (TGF- 尾 1) in human glomerular Mesangial cells (HMCs). Methods HMCs was divided into two groups: the control group with TGF- 尾 1 neutralizing antibody, the group with high glucose and high fat, the control group with TGF- 尾 1 neutralizing antibody and the group with high glucose and high fat. High glucose and high fat TGF- 尾 1 neutralizing antibody group, The expression of TGF- 尾 _ 1 mRNA in supernatant was detected by qRT-PCR and Elisa.
