去卵巢對骨質(zhì)疏松大鼠皮膚衰老的影響

【摘要】:目的用去卵巢大鼠構(gòu)建的骨質(zhì)疏松癥模型是研究絕經(jīng)后骨質(zhì)疏松疾病的最適模型,然而大鼠卵巢去除后對皮膚的影響如何,這方面的研究甚少。本研究旨在觀察去卵巢對大鼠皮膚衰老指標(biāo)的影響。方法 3月齡健康SD雌性大鼠20只,隨機(jī)分為假手術(shù)組(Sham組)和去卵巢組(OVX組),卵巢去除后3個(gè)月,測定血清雌二醇,以及皮膚組織中與衰老相關(guān)的生化指標(biāo)如過氧化氫酶(CAT)、谷胱甘肽過氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活性以及丙二醛(MDA)、羥脯氨酸(HYP)、脂褐質(zhì)(LF)含量,并觀察皮膚組織病理學(xué)變化,用計(jì)算機(jī)圖像分析系統(tǒng)定量分析。結(jié)果 OVX組大鼠血清雌二醇水平顯著降低;組織病理學(xué)觀察顯示OVX組表皮、真皮厚度較Sham組顯著變薄(P0.01),皮膚膠原纖維減少,排列疏松;與Sham組相比,OVX組大鼠皮膚CAT、GSH-Px和SOD活性顯著降低(P0.01),MDA含量較Sham組明顯增加(P0.01),皮膚組織中HYP含量明顯減少,LF含量顯著增多(P0.01)。結(jié)論 3月齡大鼠卵巢去除后3個(gè)月可導(dǎo)致皮膚衰老,可能是研究絕經(jīng)后皮膚衰老的適宜模型。
[Abstract]:Objective Osteoporosis model in ovariectomized rats is the best model for studying postmenopausal osteoporosis. The purpose of this study was to observe the effect of ovariectomy on skin senescence in rats. Methods Twenty healthy female SD rats aged 3 months were randomly divided into sham-operated group (Sham group) and ovariectomized group (OVX group). Serum estradiol, and the activities of catalase CATX, glutathione peroxidase (GSH-PxX), superoxide dismutase (SOD) and the contents of malondialdehyde (MDA), hydroxyproline (HYP), lipofuscin (LFLF) in skin tissue were measured. The histopathological changes of skin were observed and quantitatively analyzed by computer image analysis system. Results the serum estradiol level in OVX group was significantly decreased, and the histopathological observation showed that the epidermis of OVX group, The thickness of dermis was significantly thinner than that of Sham group (P 0.01), the collagen fibers of skin decreased and the arrangement was loose. Compared with Sham group, the activity of GSH-Px and SOD in skin of OVX group was significantly lower than that of Sham group. Compared with Sham group, the content of HYP in skin tissue was significantly decreased, and the content of LF was significantly increased. Conclusion 3 months after the removal of ovary of 3 months old rats, skin senescence can be induced. It may be a suitable model for studying postmenopausal skin aging.
