維生素D對絕經(jīng)后骨質疏松癥基質Gla蛋白表達的影響

【摘要】:目的:1、觀察絕經(jīng)后骨質疏松患者血清基質Gla蛋白(MGP)與骨密度、血清MGP與25(OH)D3的關系;2、觀察1,25(OH)2D3對人骨肉瘤細胞MG63的Wnt信號通路及MGP表達的影響,探討1,25(OH)2D3在絕經(jīng)后骨質疏松癥防治作用中可能的新機制。方法:1、研究對象的納入:2013年4月到2015年2月南昌大學第二附屬醫(yī)院門診體檢及住院的絕經(jīng)后女性125人,按骨密度可分為骨密度正常組、低骨量組、骨質疏松組。測定血清MGP、25(OH)D3濃度,將絕經(jīng)后骨質疏松癥患者按25(OH)D3水平分為充足組、不足組、缺乏組。2、分別使用1,25(OH)2D3 10-8mol/L、DKK-1(Dickkopf-1)200 ng/ml、1,25(OH)2D3 10-8 mol/L+DKK-1 200ng/ml干預人骨肉瘤細胞MG63細胞株48小時,1%BSA(bovine serum albumin,牛血清白蛋白)+無酚紅MEM培養(yǎng)作為空白對照組,使用實時熒光定量RT-PCR及western-blot技術測定Wnt/β-catenin信號通路中相關因子β-catenin、Runx2、LRP5及MGP基因及蛋白表達的情況。結果:1、絕經(jīng)后骨質疏松癥患者低骨量組、骨質疏松組血清MGP濃度明顯低于正常組,差異有統(tǒng)計學意義(P0.05),且血清MGP與骨密度呈正相關。2、絕經(jīng)后骨質疏松癥患者的25(OH)D3缺乏組、不足組的血清MGP明顯低于充足組,差異有統(tǒng)計學意義(P0.05)且血清MGP與25(OH)D3呈正相關。3、1,25(OH)2D3上調MG63細胞中Wnt/β-catenin信號通路中相關因子β-catenin、Runx2、LRP5時,MGP的表達也增加。DKK-1抑制Wnt/β-catenin信號通路的同時下調了MGP的表達,差異均有統(tǒng)計學意義(P0.05)。結論:1、絕經(jīng)后骨質疏松癥患者血清MGP與骨密度呈正相關。2、絕經(jīng)后骨質疏松癥患者血清MGP濃度與25(OH)D3呈正相關。3、1,25(OH)2D3有可能通過Wnt/β-catenin信號通路影響MGP的表達。
[Abstract]:Objective: To observe 1 postmenopausal osteoporosis serum matrix Gla protein (MGP) and bone mineral density, serum MGP and 25 (OH) D3; 2, 1,25 (OH) to observe the influence of 2D3 expression on human osteosarcoma cell line MG63 Wnt pathway and MGP, on 1,25 (OH) 2D3 in menopausal women after the osteoporosis prevention and treatment may be a new mechanism. Methods: 1, included in the study: from April 2013 to February 2015 in the Second Affiliated Hospital of Nanchang University medical outpatient and inpatient postmenopausal women 125 people, according to the bone density, bone mineral density can be divided into normal group, osteopenia group, osteoporosis group. Serum MGP, 25 (OH) D3 concentration, the postmenopausal osteoporosis patients by 25 (OH) D3 levels were divided into deficiency group, lack of adequate group,.2 group, respectively, using 1,25 (OH) 2D3 10-8mol/L DKK-1 (Dickkopf-1) 200 ng/ml, 1,25 (OH) 2D3 10-8 mol/L+DKK-1 200ng/ml intervention in human osteosarcoma cell line MG63 cells 48 hours, 1%BSA (bovine Serum albumin, bovine serum albumin) + phenol red free MEM medium as the blank control group, real-time fluorescence quantitative RT-PCR and Western-blot determination of related factor beta Wnt/ beta -catenin signaling pathway in -catenin, Runx2, LRP5 and MGP gene and protein expression. Results: 1 postmenopausal osteoporosis patients with low bone mass group, osteoporosis osteoporosis group serum MGP concentration was significantly lower than the normal group, the difference was statistically significant (P0.05), and the serum MGP was positively correlated with bone mineral density.2, the patients with osteoporosis after menopause 25 (OH) D3 deficient group, serum MGP deficiency group was significantly lower than that in sufficient group, the difference was statistically significant (P0.05) and the serum MGP and 25 (OH) D3 was positively correlated with.3,1,25 (OH) related factor beta Wnt/ beta -catenin signaling pathway in -catenin cells, 2D3 upregulation of MG63 Runx2, LRP5, MGP also increased expression of.DKK-1 inhibits Wnt/ beta -catenin signaling pathway and down-regulation of MGP The expression, the differences were statistically significant (P0.05). Conclusion: 1 postmenopausal osteoporosis patients serum MGP was positively correlated with bone mineral density.2, postmenopausal osteoporosis in patients with serum MGP concentration (OH) and 25 D3 (OH).3,1,25 was positively related to 2D3 through Wnt/ beta -catenin signaling pathway affect the expression of MGP.
