2型糖尿病小鼠周圍神經(jīng)病變模型的建立及皮下神經(jīng)纖維密度的檢測(cè)

【摘要】:目的建立2型糖尿病小鼠周圍神經(jīng)病變模型,并對(duì)該模型的皮下神經(jīng)纖維密度(intra-epidermal nerve fiber density,IENFD)進(jìn)行檢測(cè)。方法 C57BL6雄性小鼠24只,隨機(jī)分為四組,每組6只:HS組采用高脂飼料喂養(yǎng)24周+鏈脲佐菌素STZ(120 mg/kg)單次注射;H組采用高脂飼料喂養(yǎng)24周+緩沖液注射;S組普通飼料喂養(yǎng)24周+STZ(120 mg/kg)單次注射;C組普通飼料喂養(yǎng)24周+緩沖液注射。四組小鼠均以24周為實(shí)驗(yàn)終點(diǎn),測(cè)定隨機(jī)血糖、胰島素抵抗指數(shù)(HOMA-IR)、機(jī)械痛閾和IENFD。結(jié)果第24周時(shí)HS組隨機(jī)血糖、HOMA-IR明顯高于H、S、C組(P0.01),機(jī)械痛閾明顯低于H、S、C組(P0.05),IENFD明顯低于S組和C組(P0.05);H組隨機(jī)血糖、HOMA-IR明顯高于C組(P0.01),機(jī)械痛閾與S組、C組差異無(wú)統(tǒng)計(jì)學(xué)意義,IENFD與HS組差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論高脂飼料喂養(yǎng)聯(lián)合中等劑量STZ成功建立了2型糖尿病小鼠周圍神經(jīng)病變模型,小鼠皮下神經(jīng)纖維密度明顯下降。
[Abstract]:Objective to establish a model of peripheral neuropathy in type 2 diabetic mice and to detect the subcutaneous nerve fiber density of intra-epidermal nerve fiber density.Methods Twenty-four male C57BL6 mice were randomly divided into four groups. Each group of 6 HS rats were fed high fat diet for 24 weeks with streptozotocin STZ(120 mg / kg) single injection of streptozotocin (STZ(120 mg / kg). Group H was fed with high fat feed for 24 weeks. Group S was fed with normal diet for 24 weeks. Group C was fed with normal diet for 24 weeks. After 24 weeks of culture, the mice were injected with buffer solution for 24 weeks. Random blood glucose was measured, Results at the 24th week, the HOMA-IR of HS group was significantly higher than that of P0.01C group, and the mechanical pain threshold was significantly lower than that of P0.05- IENFD in group C and P0.05H group. The HOMA-IR in HS group was significantly higher than that in group C, and the mechanical pain threshold in group C was significantly higher than that in group C (P0.05H group), and the level of HOMA-IR in group C was significantly higher than that in group C (P 0.01), and the mechanical pain threshold in group C was significantly lower than that in group C and group S (P 0.05H). There was no significant difference between group C and group S in pain threshold. There was no significant difference between IENFD and HS group. Conclusion the model of peripheral neuropathy in type 2 diabetic mice was successfully established by high fat diet and moderate dose STZ. The density of subcutaneous nerve fibers decreased significantly in mice.
