熱射病小鼠早期中樞神經(jīng)炎癥和外周炎癥的變化

【摘要】:目的探討熱射病小鼠早期(0~24 h)中樞神經(jīng)炎癥和外周炎癥的變化及其相關(guān)性。方法 70只BALB/c雄性小鼠按隨機(jī)數(shù)字表法分為正常對照組和熱射病后各時(shí)相(0、1、6、12、24 h)觀察組。應(yīng)用環(huán)境模擬艙建立熱射病小鼠模型。觀察記錄各組小鼠生存率、直腸溫度變化和體質(zhì)量變化,Real-time PCR檢測大腦皮層和海馬炎癥因子TNF-α、IL-1β和IL-6的表達(dá),ELISA測定外周血清中炎癥因子TNF-α、IL-1β和IL-6的分泌情況,鱟試劑動態(tài)濁度法檢測血清中內(nèi)毒素濃度。結(jié)果熱暴露后小鼠直腸溫度超過42℃,1~2 h出現(xiàn)低溫期,24 h后小鼠存活率為78.57%,成功制備熱射病小鼠模型;大腦皮層及海馬在熱暴露后1 h炎癥因子TNF-α、IL-1β、IL-6的基因表達(dá)顯著增加并達(dá)到最高峰值(P0.01),6 h后逐漸降低但仍明顯高于正常對照組(P0.01),24 h后基本恢復(fù)至正常對照組水平(P0.05);外周血清中炎癥因子TNF-α、IL-1β和IL-6在1 h后含量顯著增加(P0.01),之后緩慢下降,到24 h后仍高于正常對照組(P0.01);血清內(nèi)毒素在熱暴露即刻(0 h)開始明顯升高,6 h后達(dá)到峰值,至24 h仍明顯高于正常對照組(P0.01)。結(jié)論熱射病小鼠在熱暴露12 h內(nèi)中樞神經(jīng)系統(tǒng)出現(xiàn)一過性炎癥反應(yīng),而外周炎癥反應(yīng)持續(xù)至24 h之后,提示熱射病小鼠早期中樞神經(jīng)炎癥反應(yīng)與外周炎癥反應(yīng)可能是相互獨(dú)立發(fā)生。
[Abstract]:Objective to investigate the changes of central nervous system inflammation and peripheral inflammation and their correlation in mice with heat emitting disease. Methods 70 male BALB/c mice were randomly divided into two groups: normal control group and control group. Model of heat emitter mice was established by using environmental simulation module. Survival rate of mice in each group was observed and recorded. Changes of rectal temperature and body mass. Real-time PCR was used to detect the expression of TNF- 偽 and IL-1 尾 and IL-6 in cerebral cortex and hippocampus.
