慢性高原病大鼠肝組織中扭轉(zhuǎn)原腸胚形成同系物1、鐵調(diào)素表達(dá)觀察

【摘要】:目的觀察慢性高原病(CMS)大鼠肝組織中扭轉(zhuǎn)原腸胚形成同系物1(TWSG1)、鐵調(diào)素的表達(dá)變化,并探討其意義。方法 50只雄性健康大鼠由西安運(yùn)至西寧,隨機(jī)分成6組,即低氧1組、低氧2組、CMS組、常氧1組、常氧2組、常氧3組。常氧1、2、3組分別在西寧(海拔2 260 m)常規(guī)飼養(yǎng)7、15、30 d。低氧1、2組和CMS組放入低壓氧倉(cāng)模擬海拔5 000 m環(huán)境,分別飼養(yǎng)7、15、30 d。CMS組成功制作CMS模型。采用qRT-PCR法檢測(cè)肝組織中的鐵調(diào)素、TWSG1 mRNA;采用免疫組化法檢測(cè)肝組織中的TWSG1蛋白。結(jié)果低氧2組、CMS組肝組織中鐵調(diào)素mRNA相對(duì)表達(dá)量分別低于常氧2、3組,CMS組低于低氧1組(P均0.05)。低氧1組、低氧2組、CMS組肝組織中TWSG1 mRNA相對(duì)表達(dá)量分別高于常氧1、2、3組,低氧1組、低氧2組、CMS組TWSG1 mRNA相對(duì)表達(dá)量依次增高(P均0.05)。低氧1組、低氧2組、CMS組TWSG1蛋白表達(dá)陽(yáng)性分別為8、9、9例,常氧1、2、3組分別為8、8、8例。低氧2組、CMS組肝組織中TWSG1蛋白相對(duì)表達(dá)量分別高于常氧2、3組,低氧1組、低氧2組、CMS組TWSG1蛋白相對(duì)表達(dá)量依次增高(P均0.05)。結(jié)論 CMS大鼠肝組織中鐵調(diào)素表達(dá)下調(diào),TWSG1表達(dá)上調(diào);隨低氧暴露時(shí)間延長(zhǎng),機(jī)體鐵調(diào)素和TWSG1表達(dá)變化越明顯,預(yù)示CMS狀態(tài)下鐵代謝逐漸發(fā)生改變。
[Abstract]:Objective to observe the changes of the expression of TWSG1 and iron in the liver tissue of rats with chronic mountain sickness (CMSM). Methods 50 healthy male rats were randomly divided into 6 groups, namely hypoxia group 1, from Xi'an to Xining. Hypoxia group 2: CMS group, normoxic group 1, normoxic group 2, normoxic group 3. Normoxic 1o 2 group was fed in Xining (2 260 m above sea level) for 7 15 ~ 30 days respectively. Hypoxia 1 + 2 group and CMS group were placed in a low pressure oxygen chamber to simulate 5 000 m above sea level. The CMS model was successfully made by feeding 7 ~ 1530 d.CMS group. The iron modulin TWSG1 mRNAs in liver tissue were detected by qRT-PCR method, and the TWSG1 protein in liver tissue was detected by immunohistochemical method. Results the relative expression of iron modulin mRNA was detected in liver tissue of hypoxia 2 groups. The difference between CMS group and hypoxia group (P < 0.05) was lower than that in normal oxygen group (P < 0.05), while that in hypoxia group 1 was lower than that in control group (P < 0.05). The relative expression of TWSG1 mRNA in liver tissue of hypoxia 2 group was higher than that of normoxic 1 1 group, hypoxia 1 group and hypoxia 2 group (P < 0 05). The positive expression of TWSG1 protein in hypoxia group and hypoxia 2 group were 89 cases, respectively. The relative expression of TWSG1 protein in liver tissue of hypoxia group 2 was higher than that of normoxic group 2 and hypoxia group 1, respectively. Conclusion the expression of iron modulin in liver tissue of CMS rats is down-regulated and the expression of TWSG1 is up-regulated, and the changes of iron modulin and TWSG1 expression are more obvious with the prolongation of hypoxia exposure time. It is indicated that iron metabolism changes gradually in CMS state.
