苦精對(duì)哮喘小鼠氣道中鈣結(jié)合蛋白S100A9及氣道黏液分泌的影響

【摘要】:目的研究發(fā)現(xiàn)在人體肺部存在苦味受體,目前關(guān)于苦味受體激動(dòng)劑與氣道黏液分泌的研究報(bào)道較少。探討肺部苦味受體激動(dòng)劑苯甲地鈉銨(苦精)對(duì)哮喘小鼠氣道中鈣結(jié)合蛋白S100A9水平和氣道中黏液分泌的影響。方法將健康清潔型雄性BALB/c小鼠按隨機(jī)數(shù)字表分成5組:正常對(duì)照組,慢性哮喘組,苦精干預(yù)1、2、3周組,每組8只。除正常對(duì)照組外,其余組小鼠于第1天和第15天分別腹腔注射0.2 m L氫氧化鋁混懸液[含100μg OVA,1 mg Al(OH)3)]致敏?嗑深A(yù)1、2、3周組小鼠分別從第10、9、8周起在激發(fā)的基礎(chǔ)上吸入800μg苦精1、2、3周。ELISA法及免疫組化法檢測(cè)肺泡灌洗液(BALF)上清、肺組織中S100A9蛋白水平,糖原PAS染色及實(shí)時(shí)定量PCR顯示氣道杯狀細(xì)胞數(shù)量、黏液性物質(zhì)及黏蛋白MUC5AC的含量。結(jié)果與正常對(duì)照組BALF上清中S100A9水平[(6.19±0.61)ng/m L]比較,慢性哮喘組以及苦精干預(yù)1、2周組[(11.89±0.77)、(10.69±0.79)、(9.49±0.99)ng/m L]明顯升高(P0.05);苦精干預(yù)1、2、3周組較慢性哮喘組降低(P0.05)。與正常對(duì)照組比較,免疫組化染色顯示慢性哮喘組氣道上皮細(xì)胞中S100A9高表達(dá),苦精干預(yù)1、2、3周后,氣道上皮細(xì)胞中S100A9表達(dá)隨苦精干預(yù)時(shí)間增加而減少。糖原PAS染色顯示,與正常對(duì)照組比較,慢性哮喘組氣道上皮杯狀細(xì)胞增生,黏液過(guò)度分泌(P0.01)。與慢性哮喘組比較,苦精干預(yù)1、2、3周組杯狀細(xì)胞數(shù)量、黏液分泌量隨苦精干預(yù)時(shí)間的增加而減少。與正常對(duì)照組比較,慢性哮喘組以及苦精干預(yù)1、2周組肺組織中黏蛋白MUC5AC mRNA表達(dá)增高,差異有統(tǒng)計(jì)學(xué)意義(P0.05),與慢性哮喘組比較,苦精干預(yù)后,MUC5AC的mRNA表達(dá)降低,并隨苦精干預(yù)時(shí)間的增加而降低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論苦精下調(diào)小鼠杯狀細(xì)胞的黏液分泌可能與上皮細(xì)胞鈣結(jié)合蛋白S100A9有關(guān)。
[Abstract]:Objective to study the existence of bitter receptors in human lungs. There are few reports about bitter receptor agonists and airway mucus secretion. To explore the effects of pulmonary bitter receptor agonist benzodimethylammonium (bitter essence) on the levels of calcium binding protein S100A9 in the airway and mucus secretion in the airway of asthmatic mice. Methods healthy and clean male BALB/c mice were randomly divided into 5 groups: normal control group. In the chronic asthma group, 8 rats in each group were treated with bitter extract for 3 weeks, except for the normal control group. The mice in the other group were sensitized by intraperitoneal injection of 0.2 mL aluminum hydroxide suspension (containing 100 渭 g OVAX 1 mg Albumin 3) on day 1 and day 15, respectively. And immunohistochemical method was used to detect the supernatant of alveolar lavage fluid (BALF). The levels of S100A9 protein in lung tissue, glycogen PAS staining and real-time quantitative PCR showed the number of goblet cells, mucin and mucin MUC5AC in the airway. The results were compared with those in the normal control group (6.19 鹵0.61g / mL). The expression of S100A9 in airway epithelial cells of chronic asthma group was significantly higher than that of chronic asthma group after 1 ~ 2 weeks of treatment [11.89 鹵0.77 ~ 10.69 鹵0.79 ng / mL], and the expression of S100A9 in airway epithelial cells of chronic asthma group was significantly higher than that of chronic asthma group at 3 weeks after treatment with bitter spermatine for 1 ~ 2 ~ 2 weeks. Compared with normal control group, the expression of S100A9 in airway epithelial cells in chronic asthma group was significantly higher than that in control group, and the expression of S100A9 in airway epithelial cells in chronic asthma group was significantly higher than that in control group. The expression of S100A9 in airway epithelial cells decreased with the increase of the time of intervention. Glycogen PAS staining showed that, compared with the normal control group, the airway epithelial goblet cells proliferated in chronic asthma group. Compared with the chronic asthma group, the number of goblet cells in the group treated with bitter spermatine for 3 weeks was decreased with the increase of the time of the intervention. Compared with the normal control group, the number of goblet cells decreased with the increase of the time of the treatment. The expression of mucin MUC5AC mRNA in the lung tissue of chronic asthma group and the group treated with bitter essence for 1 to 2 weeks was increased, the difference was statistically significant (P 0.05). Compared with the group of chronic asthma, the expression of MUC5AC mRNA in bitter dry prognosis was lower than that in chronic asthma group, and the expression of MUC5AC decreased with the increase of the time of intervention. Conclusion the down-regulation of mouse goblet cell mucus secretion may be related to the calcium binding protein S100A9.
