碘過量不同處理時間對Fischer大鼠甲狀腺細胞線粒體過氧化損傷的影響

【摘要】:目的探討碘過量不同處理時間對Fischer大鼠甲狀腺細胞線粒體過氧化損傷的影響。方法采用含0(對照組)、0.1 mmol/L碘化鉀的Coons F12培養(yǎng)基培養(yǎng)Fischer大鼠甲狀腺細胞2、6 h、1 d,對小鼠抗大鼠細胞色素C蛋白、死亡細胞百分比、線粒體超氧化物生成、乳酸脫氫酶活力進行檢測分析研究。結(jié)果 0.1 mmol/L碘化鉀組處理2 h、6 h、1 d時,小鼠抗大鼠細胞色素C強陽性細胞率遠高于對照組(P0.05);Fischer大鼠甲狀腺細胞線粒體超氧化物生成明顯高于對照組(P0.05),且處理2 h時增長幅度最大;隨著0.1 mmol/L碘化鉀處理時間的增加,培養(yǎng)液上清中乳酸脫氫酶活力程序持續(xù)增加趨勢,處理2、6 h、1 d時乳酸脫氫酶活力明顯高于對照組(P0.05)。結(jié)論碘過量處理2 h可能會導(dǎo)致Fischer大鼠甲狀腺細胞線粒體發(fā)生過氧化損傷,而處理1 d則極有可能導(dǎo)致細胞凋亡。
[Abstract]:Objective to investigate the effect of iodine excess treatment on the mitochondrial peroxidation injury of thyroid cells in Fischer rats. Methods the thyroid cells of Fischer rats were cultured on Coons F12 medium containing 0 (control group) 0.1 mmol/L potassium iodide for 2 hours for 1 day. Anti rat cytochrome C protein, The percentage of dead cells, the formation of superoxide in mitochondria and the activity of lactate dehydrogenase were detected and analyzed. Results the results showed that the 0.1 mmol/L potassium iodide group was treated with potassium iodide for 2 h and 6 h for 1 day. The rate of anti-cytochrome C strong positive cells in mice was much higher than that in control group P0.05 Fischer rats, and the production of mitochondrial superoxide in thyroid cells was significantly higher than that in control group P0.05, and the maximal increase was observed at 2 h after treatment, with the increase of 0.1 mmol/L potassium iodide treatment time. The activity of lactate dehydrogenase in the supernatant of culture medium continued to increase, and the activity of lactate dehydrogenase in the supernatant was significantly higher than that in the control group at 2 h after treatment for 6 h. Conclusion iodine excess treatment for 2 h may cause peroxidation damage to the mitochondria of thyroid cells in Fischer rats. On the other hand, the treatment for 1 day is likely to lead to apoptosis.
