miRNA-467b在EAE中對炎性T細胞浸潤的影響

【摘要】:為探討miRNA-467b在實驗性自身免疫性腦脊髓膜炎(experimental autoimmune encephalomyelitis,EAE)中對炎性T細胞浸潤的影響,我們首先采用real-time PCR及FACS方法檢測EAE小鼠脾臟單個核細胞中CXCR3、CCR5、CCR2、CCR6的mRNA和蛋白表達水平,并在MOG反應性T細胞中過表達miRNA-467b,FACS方法檢測趨化因子受體CXCR3、CCR6的表達變化,并進一步通過Boyden小室(transwell實驗)的方法,體外觀察miRNA-467b對細胞遷移的影響。結(jié)果發(fā)現(xiàn),EAE小鼠脾臟中CCR5、CCR2、CCR6的轉(zhuǎn)錄及蛋白表達水平顯著上升,在MOG特異性的T細胞中過表達miRNA-467b后,可降低Th1表面趨化因子受體的表達,減少Th1向transwell板下室中的遷移,提示miRNA-467b可以通過影響細胞的遷移進而調(diào)節(jié)EAE的疾病進程。
[Abstract]:To investigate the effect of miRNA-467b on inflammatory T cell infiltration in experimental autoimmune meningitis autoimmune encephalomytis (EAE), we first detected the expression of mRNA and protein in the spleen mononuclear cells of EAE mice by real-time PCR and FACS. The overexpression of miRNA-467bFACS in MOG reactive T cells was used to detect the expression of chemokine receptor (CXCR3) CCR6, and the expression of CCR6 was detected by Boyden transwell assay. The effect of miRNA-467b on cell migration was observed in vitro. The results showed that the expression of CCR5, CCR2CCR6 and CCR6 protein increased significantly in the spleen of MOG mice. After overexpression of miRNA-467b in MOG specific T cells, the expression of chemokine receptor on Th1 surface was decreased. Reducing the migration of Th1 to the subplate chamber of transwell suggests that miRNA-467b can regulate the progression of EAE by affecting cell migration.
