絲膠蛋白對鏈脲佐菌素誘導(dǎo)胰島細(xì)胞株INS-1凋亡相關(guān)因子的影響研究

【摘要】:目的探討絲膠蛋白對鏈脲佐菌素(STZ)誘導(dǎo)胰島細(xì)胞株INS-1凋亡相關(guān)因子B淋巴細(xì)胞瘤2(Bcl-2)、Bcl-2相關(guān)X蛋白(Bax)的影響。方法 2015年6月—2016年3月,將大鼠胰島細(xì)胞株INS-1分為3組:正常組胰島細(xì)胞株INS-1不作任何處理,STZ組以STZ作用胰島細(xì)胞株INS-1,絲膠蛋白組以STZ和絲膠蛋白作用胰島細(xì)胞株INS-1。CCK-8法檢測各組細(xì)胞存活率;流式細(xì)胞術(shù)檢測各組細(xì)胞凋亡率;實(shí)時(shí)熒光定量聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)法和Western blotting法檢測凋亡相關(guān)因子Bcl-2和Bax mRNA表達(dá)及其蛋白的表達(dá)。結(jié)果 STZ組胰島細(xì)胞株INS-1細(xì)胞存活率較正常組降低,絲膠蛋白組胰島細(xì)胞株INS-1細(xì)胞存活率較STZ組升高(P0.05)。流式細(xì)胞術(shù)檢測結(jié)果顯示,STZ組胰島細(xì)胞株INS-1細(xì)胞凋亡率較正常組升高,絲膠蛋白組胰島細(xì)胞株INS-1細(xì)胞凋亡率較STZ組降低(P0.05)。STZ組胰島細(xì)胞株INS-1 Bcl-2 mRNA及其蛋白表達(dá)水平、Bcl-2 mRNA/Bax mRNA、Bcl-2/Bax較正常組降低,Bax mRNA及其蛋白表達(dá)水平較正常組升高(P0.05);絲膠蛋白組胰島細(xì)胞株INS-1 Bcl-2 mRNA及其蛋白表達(dá)水平、Bcl-2mRNA/Bax mRNA、Bcl-2/Bax較STZ組升高,Bax mRNA及其蛋白表達(dá)水平較STZ組降低(P0.05)。結(jié)論絲膠蛋白可抑制STZ誘導(dǎo)的胰島細(xì)胞株INS-1的凋亡,提高Bcl-2/Bax的比值。
[Abstract]:Objective to investigate the effect of sericin on streptozotocin (STZ) -induced pancreatic islet cell line INS-1 apoptosis-related factor B lymphocytoma (Bcl-2). The effect of Bcl-2 related X protein Bax.Methods from June 2015 to March 2016. Rat islet cell line INS-1 was divided into three groups: normal islet cell line INS-1 was treated with STZ without any treatment and islet cell line INS-1 was treated with STZ. STZ and sericin treated islet cell line INS-1.CCK-8 were used to detect the survival rate of each group. Apoptosis rate was detected by flow cytometry. Real-time quantitative polymerase chain reaction (RT-PCR). The expression of apoptosis-related factors Bcl-2 and Bax mRNA and its protein expression were detected by Western blotting method. The survival rate of INS-1 cells in STZ group was lower than that in normal group. The survival rate of INS-1 cells in sericin group was higher than that in STZ group (P 0.05). The apoptosis rate of INS-1 cells in STZ group was higher than that in normal group. The apoptotic rate of INS-1 cells in sericin group was lower than that in STZ group. The expression level of INS-1 Bcl-2 mRNA and its protein in islet cell line INS-1 Bcl-2 in sericin group was lower than that in STZ group. Bcl-2 mRNA/Bax mRNAs Bcl-2 / Bax decreased the expression level of Bax mRNA and its protein in Bcl-2 / Bax group compared with the normal group, and the expression level of Bcl-2 / Bax protein was higher than that of the normal group. The expression level of INS-1 Bcl-2mRNA and its protein in sericin group islet cell line was determined by Bcl-2mRNA-Bax mRNA. Bcl-2/Bax was higher than that in STZ group. The expression level of Bax mRNA and its protein was lower than that of STZ group. Conclusion sericin protein can inhibit the apoptosis of islet cell line INS-1 induced by STZ. The ratio of Bcl-2/Bax was increased.
