通過生物信息學(xué)方法鑒定PTPRR和JAG1為去勢(shì)抵抗性前列腺癌(CRPC)的關(guān)鍵基因(英文)

目的:鑒定去勢(shì)抵抗性前列腺癌(CRPC)的關(guān)鍵基因。創(chuàng)新點(diǎn):(1)結(jié)合多個(gè)數(shù)據(jù)庫(kù)數(shù)據(jù),運(yùn)用生物信息學(xué)方法鑒定CRPC的關(guān)鍵基因;(2)首次報(bào)道PTPRR可能在CRPC里起關(guān)鍵作用。方法:通過下載三個(gè)GEO數(shù)據(jù)庫(kù)的m RNA微陣列數(shù)據(jù),分析CRPC和激素敏感前列腺癌之間的基因差異,對(duì)篩選出的差異基因進(jìn)行功能富集分析和蛋白質(zhì)間相互作用分析,最終篩選出兩個(gè)有重要功能的差異基因(PTPRR和JAG1)。通過在多個(gè)其他數(shù)據(jù)庫(kù)中進(jìn)行表達(dá)量驗(yàn)證和生存分析,進(jìn)一步證明這些基因的重要作用。結(jié)論:PTPRR和JAG1在CRPC中顯著增高,并與預(yù)后差相關(guān)。因此,這兩個(gè)基因有可能作為CRPC的診斷和預(yù)后的生物標(biāo)志物。
