高濃度葡萄糖對膀胱癌T24細(xì)胞增殖及β-catenin表達(dá)的影響

【摘要】:目的探討高濃度葡萄糖對膀胱癌T24細(xì)胞增殖的影響,并探討其可能的作用機(jī)制。方法用含10%FBS的DMEM培養(yǎng)液體外培養(yǎng)膀胱癌T24細(xì)胞,將細(xì)胞分為5組:空白組(5.5 mmol/L葡萄糖)、對照組(5.5 mmol/L葡萄糖+25 mmol/L甘露醇)、高糖組(10、20、30 mmol/L葡萄糖)。采用MTT法及克隆形成實(shí)驗(yàn)檢測高濃度葡萄糖對膀胱癌T24細(xì)胞增殖的影響;通過實(shí)時定量PCR檢測膀胱癌T24細(xì)胞β-catenin mR NA的表達(dá)水平;通過Western blot法檢測高濃度葡萄糖對膀胱癌T24細(xì)胞β-catenin蛋白表達(dá)的影響。結(jié)果 MTT法及克隆形成實(shí)驗(yàn)結(jié)果顯示,高濃度葡萄糖顯著促進(jìn)膀胱癌T24細(xì)胞的增殖(P0.01);Western blot檢測結(jié)果顯示,高濃度葡萄糖促進(jìn)T24細(xì)胞β-catenin蛋白的表達(dá)。實(shí)時定量PCR檢測結(jié)果顯示,高濃度葡萄糖促進(jìn)T24細(xì)胞β-catenin mR NA的表達(dá)。結(jié)論高濃度葡萄糖明顯促進(jìn)膀胱癌T24細(xì)胞的增殖,并促進(jìn)T24細(xì)胞β-catenin蛋白和其mRNA的表達(dá),該作用機(jī)制可能與β-catenin表達(dá)上調(diào)有關(guān)。

[Abstract]:Objective To study the effect of high-concentration glucose on the proliferation of bladder cancer T24 cells and to explore its possible mechanism of action. Methods T24 cells of bladder cancer were cultured with DMEM containing 10% FBS, and the cells were divided into 5 groups: blank group (5.5 mmol/ L glucose), control group (5.5 mmol/ L glucose + 25 mmol/ L mannitol) and high-sugar group (10,20,30 mmol/ L glucose). The effect of high concentration of glucose on the proliferation of bladder cancer T24 cells was measured by MTT and clone formation. The expression level of T24 cells in bladder cancer T24 cells was detected by real-time quantitative PCR. The effect of high concentration of glucose on the expression of antigen-cata protein in bladder cancer T24 cells was detected by Western blot. Results The results of MTT and clone formation showed that high-concentration glucose significantly promoted the proliferation of T24 cells in bladder cancer (P0.01), and the results of Western blot showed that high-concentration glucose promoted the expression of T-cata protein in T24 cells. The results of real-time quantitative PCR showed that high-concentration glucose promoted the expression of T24 cell-cattenin mRNA. Conclusion High-concentration glucose can promote the proliferation of T24 cells in bladder cancer and promote the expression of T24 cell-cattenin protein and its mRNA. in-house picture: image indicate that high-concentration glucose promote proliferation of T24 cells of bladder cancer: control group; B: blank group; C:10 mmol/ L high-sugar group; D:20 mmol/ L high-sugar group; E:30 mmol/ L high-sugar group
